摘  要:目的 探索ZC3HAV1在多形性胶质母细胞瘤(GBM)中的表达及作用。方法 收集35例于2018年1月-2020年10月于河北医科大学第二医院手术治疗的胶质母细胞瘤患者肿瘤标本(肿瘤组)及同期重度颅脑创伤患者正常脑组织标本(对照组)20例,利用Western blot与PCR方法检测两组ZC3HAV1的表达水平;通过慢病毒转染U87细胞下调ZC3HAV1的表达水平,再利用CCK、Transwell与流式测凋亡实验验证ZC3HAV1低表达对GBM细胞增殖、侵袭与凋亡的作用;利用PCR检测35例肿瘤组织中SNHG5与ZC3HAV1的表达水平并分析两者表达的相关性;通过pcDNA及siRNA转染U87细胞上调或下调中SNHG5的表达水平,再利用Western blot检测ZC3HAV1的表达水平的变化来探索在GBM中SNHG5对ZC3HAV1表达的影响。结果 肿瘤组中ZC3HAV1的表达水平显著高于对照组(P<0.05);下调U87细胞中ZC3HAV1的表达水平可有效抑制U87细胞的增殖、侵袭并促进其细胞凋亡。GBM标本组织中SNHG5与ZC3HAV1的表达水平呈正相关,抬高U87细胞中SNHG5表达后3HAV1ZC的表达水平显著上升,下调U87细胞中SNHG5表达后ZC3HAV1表达水平明显下降(P<0.05)。结论 ZC3HAV1在多形性胶质母细胞瘤呈显著高表达并促进肿瘤的发生与进展,在胶质母细胞瘤中SNHG5可正向调控ZC3HAV1的表达。Objective To explore the expression and role of ZC3 HAV1 in glioblastoma multiform(GBM).Method The tumor samples(tumor group) and non-neoplasm brain tissue(control group) were collected intraoperatively from 35 cases of GBM and 20 patients with severe brain injury in the Second Hospital of Hebei Medical University from January 2018 and October 2020.Using qRT-PCR and Western blot tests,the difference of ZC3 HAV1 expression between two groups was determined.The downregulation of ZC3 HAV1 inU87 cell was achieved after transfection with decreasing ZC3 HAV1 lentivirus.CCK,Transwell assay and flow cytometry tests were performed to identify the role of ZC3 HAV1 in the proliferation,invasion and apoptosis of U87 cells.The expression correlation between SNHG5 and ZC3 HAV1 in GBM sample was analyzed using PCR tests.Utilizing Western Blot tests,the regulatory effect of SNHG5 on ZC3 HAV1 expression in U87 was investigated following transfection of U87 cells with pcDNA and siRNA of SNHG5.Result The expression levels of ZC3 HAV1 mRNA and protein were higher in GBM tissues than those in normal brain tissues(P<0.05).In vitro,decreased ZC3 HAV1 inhibited effectively the proliferation and invasion,as well as promoted the apoptosis of U87 cells.ZC3 HAV1 expression correlated positively with SNHG5 expression in GBM.ZC3 HAV1 expression was upregulated following increasing SNHG5 expression in U87 cells,ZC3 HAV1 expression was decreased after downregulation of SNHG5 in U87 cells.Conclusion ZC3 HAV1 expression was significantly upregulated in glioblastoma multiform and ZC3 HAV1 promoted the development and growth of GBM.SNHG5 regulated positively expression of ZC3 HAV1 in GBM.
