摘  要:目的 探讨运动神经元病相关蛋白p150glued在小鼠脊髓白质轴突变性中的作用。方法使用蛋白印迹法检测p150glued在小鼠中枢神经系统中蛋白表达水平,使用免疫荧光法检测p150glued缺失对小鼠脊髓白质中神经纤维(轴突)的影响。结果与对照组相比,实验组中p150glued在小鼠脊髓神经元中蛋白质含量显著下降;与对照组相比,实验组中小鼠脊髓(颈椎)皮质脊髓束中轴突数量变化不明显,而脊髓小脑束中直径较大轴突数量显著下降。结论p150glued缺失可导致迟发的小鼠脊髓白质中发生特异性轴突变性。Objective To investigate the effects of motor neuron disease associated protein p150 glued in axonal degeneration of mouse spinal cord white matter.Methods The expression of p150 glued in the central nervous system of mouse were examined by Western blot.The role of p150 glued knock down in nerve fibers(axons) in the corticospinal tract and the spinocerebellar tract of mouse spinal cord white matter were examined by immunofluorescence and laser confocal scanning microscopy.Results Compared to control group,the experimental group showed decreased level of p150 glued in spinal cord,and the number of axons showed comparable level in the corticospinal tract,the number of axons with larger diameter decreased significantly in the spinocerebellar tract.Conclusion p150 glued deficiency can lead to delayed and specific axonal degeneration in mouse spinal cord white matter.
