摘　　要：目的:探究上调胰岛素样生长因子结合蛋白6(IGFBP-6)基因对乳腺癌细胞MDA-MB-231细胞周期和凋亡的影响。方法:体外培养人三阴性乳腺癌MDA-MB-231细胞,转染IGFBP-6过表达质粒,实时荧光定量PCR法验证转染效率;流式细胞术检测转染IGFBP-6后的细胞周期的改变及凋亡的情况;免疫印迹检测增殖细胞核抗原(PCNA)、细胞周期蛋白D1(cyclin D1)、凋亡相关蛋白Bcl-2和Bax的蛋白表达水平。结果:IGFBP-6转染MDA-MB-231细胞后,IGFBP-6的mRNA水平明显上调;PCNA、细胞周期蛋白D1的表达水平明显降低;细胞G1/S期阻滞;凋亡细胞增多。结论:IGFBP-6可以显著抑制乳腺癌细胞的增殖,促进细胞凋亡。Objective:To investigate the effects of up-regulation of insulin-like growth factor-binding protein 6(IGFBP-6) gene on the MDA-MB-231 cycle and apoptosis in breast cancer cells.Methods:Human triple negative breast cancer cells MDA-MB-231 were cultured in vitro and transfected with IGFBP-6 overexpressing plasmid.The transfection efficiency was verified by real-time quantitative PCR.The change of IGFBP-6 cell cycle and apoptosis after transfection were detected by flow cytometry.The protein expression levels of proliferating nuclear antigen(PCNA),cyclin D1,Bcl-2 and Bax were detected by Western blotting.Results:After transfection of MDA-MB-231 cells,the mRNA level of IGFBP-6 was significantly up-regulated,the expression levels of PCNA and cyclin D1 were significantly decreased,G1/S phase arrest,and the apoptotic cells were increased.Conclusion:IGFBP-6 can significantly inhibit the proliferation and promote apoptosis of breast cancer cells.
