摘　　要：目的:探讨脑出血对酵母沉默信息调节因子2(Sirt2)和炎症的影响。方法:将胶原酶Ⅳ注入SD大鼠右侧纹状体中建立脑出血模型,通过免疫印迹和ELISA等方法测定大鼠脑出血后48 h的Sirt2的表达及炎症变化。利用Hemin诱导PC12细胞损伤模拟体外脑出血模型,并检测Sirt2及炎症变化;采用短发夹RNA(shRNA)-Sirt2沉默Sirt2在PC12细胞中的表达及对炎症的影响。结果:手术后48 h脑出血行为学评分最低。脑出血组Sirt2的表达显著高于假手术组。脑出血组IL-6、IL-1β表达显著升高。结论:脑出血可以促进Sirt2的表达和炎症反应,降低Sirt2的表达可减缓炎症反应。Objective:To investigate the effect of intracerebral hemorrhage(ICH) on yeast silent information regulator 2(Sirt2) and inflammation.Methods:Collagenase Ⅳ was injected into the right striatum of SD rats to establish a rat ICH model.The expression of Sirt2 and inflammatory changes in rats at 48 h after ICH were measured by Western botting and ELISA.Hemin was used to induce PC12 cell damage to simulate the in vitro ICH model and detect Sirt2 and inflammation changes.Short hairpin RNA(shRNA)-Sirt2 was used to silence the expression of Sirt2 in PC12 cells and its influence on inflammation.Results:The behavioral score of cerebral hemorrhage was the lowest at 48 hours after surgery.The expression of Sirt2 in the ICH group was significantly higher than that in the sham operation group.The expression of IL-6 and IL-1β in ICH group was significantly increased.Conclusion:ICH can promote the expression of Sirt2 and inflammatory response,and reducing the expression of Sirt2 can slow down the inflammatory response.
