摘 要:目的基于cAMP/PKA信号通路探讨通脉养心丸对缓慢性心律失常的干预作用及机制。方法将40只健康雄性SD大鼠随机分为空白组、模型组、通脉养心低剂量组、通脉养心高剂量组,每组10只。空白组大鼠实验全程灌胃生理盐水;模型组大鼠上午灌胃盐酸普萘洛尔诱发缓慢性心律失常,下午灌胃生理盐水;通脉养心低、高剂量组上午灌胃盐酸普萘洛尔诱发缓慢性心律失常,下午分别灌胃通脉养心丸2 g/kg和4 g/kg。各组均连续灌胃4周。测定各组大鼠干预前及干预2周、干预4周后心率;处死各组大鼠,ELISA法测定心肌组织中cAMP、PKA含量,PCR法和Western blot法分别检测心肌组织中cAMP、PKA、L型钙通道α1C亚基(CACNA1C)mRNA及蛋白表达情况。结果干预2周和4周后模型组大鼠心率均明显慢于同期空白组(P均<0.05),通脉养心低、高剂量组均明显快于同期模型组(P均<0.05),且干预4周后通脉养心高剂量组大鼠心率明显快于通脉养心低剂量组(P<0.05)。模型组大鼠心肌组织中cAMP、PKA含量均明显低于空白组(P均<0.05);通脉养心低、高剂量组大鼠心肌组织中cAMP含量及通脉养心高剂量组大鼠心肌组织PKA含量均明显高于模型组(P均<0.05),通脉养心低、高剂量组大鼠心肌组织中cAMP、PKA含量比较差异均无统计学意义(P均>0.05)。模型组大鼠心肌组织中cAMP、PKA、CACNA1C mRNA及蛋白相对表达量均明显低于空白组(P均<0.05);通脉养心高剂量组大鼠心肌组织中cAMP、PKA、CACNA1C mRNA及蛋白相对表达量均明显高于模型组(P均<0.05),通脉养心低剂量组仅cAMP蛋白相对表达量明显高于模型组(P<0.05);通脉养心高剂量组大鼠心肌组织中cAMP、PKA、CACNA1C mRNA及蛋白相对表达量与通脉养心低剂量组比较差异均无统计学意义(P均>0.05)。结论通脉养心丸能够提升缓慢性心律失常大鼠心率,其作用机制可能与激活cAMP/PKA信号通路进而磷酸化L型钙通�Objective It is to observe the effects Tongmai Yangxin pill on heart rate of rats with bradyarrhythmia based on cAMP/PKA signaling pathway.Methods Forty healthy male SD rats were randomly divided into blank group,model group,low-dose Tongmai Yangxin pill group and high-dose Tongmai Yangxin pill group,with 10 rats in each group.The rats in the blank group were given normal saline by gavage throughout the experiment,the rats in the model group were treated with propranolol hydrochloride by gavage in the morning to induce bradyarrhythmias,and treated with normal saline by gavage in the afternoon,the low-dose and high-dose Tongmai Yangxin groups were given propranolol hydrochloride by gavage to induce bradyarrhythmias in the morning,and respectively given Tongmai Yangxin pills 2 g/kg and 4 g/kg by gavage in the afternoon.All groups were continuously treated for 4 weeks.The heart rate of the rats in each group was detected before the intervention,after 2 weeks and 4 weeks of the intervention.At the end of the experiment,the rats in each group were sacrificed,the contents of cAMP and PKA in myocardial tissue were detected by ELISA,the expression of mRNA and protein of cAMP,PKA and L-type calcium channelα1C subunit(CACNA1C)in myocardial tissue were respectively detected by PCR and Western blot.Results After 2 weeks and 4 weeks of intervention,the heart rate of rats in the model group was significantly slower than that in the blank group(P<0.05),and the rate of the rats in the low and high dose groups of Tongmai Yangxin pill were significantly faster than that in the model group(P<0.05),and the heart rate of the rats in the high-dose Tongmai Yangxin pill group was significantly faster than that in the low-dose Tongmai Yangxin pill group after 4 weeks of intervention(P<0.05).The contents of cAMP and PKA in the myocardial tissue of the rats in the model group were significantly lower than those in the blank group(all P<0.05),the content of cAMP in the myocardial tissue of the rats in the low-dose and high-dose Tongmai Yan
