摘  要:目的:研究表皮生长因子受体反馈抑制剂1(ERRFI1)在癫痫小鼠海马中的表达变化,并且进行细胞定位,为探索ERRFI1在癫痫发生和发展中的作用及其机制奠定基础。方法:选取6~8周的健康雌性昆明小鼠,建立癫痫的匹罗卡品模型,采用real time RT-PCR和Western Blot分别检测对照组和各癫痫组小鼠海马中ERRFI1 mRNA和蛋白质的表达情况,用免疫荧光组织化学技术观察ERRFI1在对照组和各癫痫组小鼠海马中神经元、星形胶质细胞以及小胶质细胞的表达情况。结果:ERRFI1 mRNA在海马中的表达于癫痫持续状态(SE)1 h和2 h时显著上调(P<0.001);ERRFI1蛋白质的表达稍滞后于mRNA表达,在SE 6 h、10 h、1 d及2 d时显著上调(P<0.01)。免疫荧光结果显示,ERRFI1在海马的神经元和星形胶质细胞中表达;SE 1 d和SE 2 d时,ERRFI1在海马星形胶质细胞中的表达上调(P<0.001),在神经元中的表达无显著差异(P>0.05)。而在对照组和癫痫组小鼠的海马中,小胶质细胞都不表达ERRFI1。结论:ERRFI1通过调控海马中星形胶质细胞的功能而不是神经元和小胶质细胞的功能来参与癫痫的病理过程。Objective:To lay a foundation for exploring the role and mechanism of ERRFI1 in the occurrence and development of epilepsy,the expression changes and cellular localization of ERRFI1 were investigated in the hippocampi of epileptic mice.Methods:The pilocarpine model of epilepsy was established by using healthy female Kunming mice of 6~8 weeks old.After harvesting hippocampi of mice,the mRNA levels of ERRFI1 were detected by real-time RT-PCR and protein levels were detected by Western Blot in the hippocampi of the controlled and epileptic mice.The expression of EERRFI1 in neurons,astrocytes,and microglia was examined by immunofluorescent co-localization assay in the hippocampi of the controlled and epileptic mice.Results:The mRNA levels of ERRFI1 were significantly upregulated at SE 1 h and SE 2 h(P<0.001).The significant increase in protein levels were detected at SE 6 h,10 h,1 d,and 2 d subsequently(P<0.01).Immunofluorescent staining revealed that ERRFI1 proteins localized in hippocampal neurons and astrocytes.ERRFI1 proteins were upregulated in hippocampal astrocytes at SE 1 d and SE 2 d(P<0.001),and no significant expression change of proteins was detected in neurons(P>0.05).However,ERRFI1 immunopositive microglia was not observed in in the hippocampi of the controlled and epileptic mice.Conclusion:ERRFI1 is involved in the pathological process of epilepsy by regulating the function of astrocytes rather than neurons and microglia in the hippocampi.
