摘 要:目的:研究线粒体靶向抗氧化剂MitoQ对蛛网膜下腔出血(SAH)模型大鼠氧化应激的作用及分子机制。方法:雄性SD大鼠分为假手术组(sham)、SAH模型组和不同剂量MitoQ处理组。利用血管穿孔法制备SAH大鼠模型,MitoQ组大鼠模型制备后通过腹腔分别注射1、5和10 mg/kg MitoQ处理。利用改良的Garcia实验和平衡木实验(BBT)检测大鼠的神经功能改变,利用商品化试剂盒检测线粒体ROS水平、细胞内丙二醛(MDA)和ATP变化。结果:和假手术组大鼠相比,SAH术后48 h,5 mg/kg MitoQ处理可明显改善SAH大鼠神经功能(P<0.05),降低线粒体ROS的含量,降低脑组织中MDA的水平,同时增加脑组织ATP的水平(P<0.05)。结论:MitoQ通过减轻蛛网膜下腔出血大鼠线粒体氧化应激反应发挥保护作用。Objective:To study the effect and molecular mechanism of mitochondrial targeted antioxidant MitoQ on oxidative stress in rats with subarachnoid hemorrhage(SAH).Methods:Male SD rats were divided into sham operation group(sham),SAH model group and MitoQ treatment group with different doses.SAH rat model was prepared by endovascular perforation.and MitoQ rat model was prepared by intraperitoneal injection of 1,5 and 10 mg/kg MitoQ,respectively.The changes of neural function were detected by modified Garcia test and balance beam test,and the changes of mitochondrial reactive oxygen species(ROS)level,intracellular malondialdehyde(MDA)and adenosine triphosphate(ATP)were detected by commercial kits.Results:Compared with sham operation group,48 h after SAH,5 mg/kg MitoQ could significantly improve the neurological function of SAH rats(P<0.05),reduce the content of mitochondrial ROS,decrease the level of MDA in brain tissue,and increase the level of ATP in brain tissue(P<0.05).Conclusion:MitoQ plays a protective role by alleviating mitochondrial oxidative stress response in rats with subarachnoid hemorrhage.
