摘 要:目的:比较研究顺铂、红景天苷对卵巢癌细胞SKOV3增殖的作用影响.方法:设置顺铂组、红景天苷组和空白组,培养SKOV3细胞至对数期,将不同浓度的顺铂和红景天苷分别作用细胞24 h、48 h和72 h,采用CCK-8法测定细胞OD值,计算药物对SKOV3细胞增殖的抑制率、半抑制浓度(IC_(50)),比较药物对SKOV3细胞的抑制率、半数抑制浓度.结果:顺铂、红景天苷对SKOV3细胞增值的抑制作用均呈浓度和作用时间的依赖性,即抑制率与药物浓度呈线性关系.两组药物对SKOV3细胞的抑制率与空白组比较差异极显著(P<0.01).顺铂对SKOV3细胞的抑制效果显著高于红景天苷;顺铂和红景天苷48 h和72 h的半数抑制浓度极显著低于24 h(P<0.01),72 h的半数抑制浓度极显著低于48 h.结论:顺铂、红景天苷对SKOV3细胞增殖的抑制率与药物浓度和作用时间呈依懒性,两药物组的IC_(50)均与药物作用SKOV3细胞时间呈依懒性,两药物的总体抑制效果相似.本研究可进一步开发红景天苷,从而为抗卵巢癌新药提供思路.Objective Comparative study of cisplatin and salidroside effects on the proliferation of ovarian cancer cells SKOV3.Methods The cisplatin group,the salidroside group and the blank group were set.SKOV3 cells were cultured to the logarithmic phase,and different concentrations of cisplatin and salidroside were added into the media at 24 h,48 h and 72 h,respectively,and the cell OD was determined by CCK-8 method.The inhibitory rate and half inhibitory concentration(IC_(50))of the drug on SKOV3 cell proliferation were calculated.The inhibitory rate and IC_(50) of the drug on SKOV3 cells were compared.Results The inhibitory effects of cisplatin and salidroside on the proliferation of SKOV3 cells were both concentration-and time-dependent,that is,the inhibition rate was linearly related to the drug concentration.The inhibitory rate of SKOV3 cells between the two groups was significantly different from that of the blank group(P<0.01).The inhibitory effect of cisplatin on SKOV3 cells was significantly higher than that of salidroside at 48h and 72h.50%inhibitory concentration was significantly lower than that at 24 h(P<0.01),and IC_(50) at 72 h was significantly lower than that of 48 h.Conclusion The inhibitory rate of cisplatin and salidroside on SKOV3 cell proliferation was dependent on drug concentration and duration of action.IC_(50) of both drug groups were also dependent on duration of drug action on SKOV3 cells,and the overall inhibitory effects of the two drugs were similar.It is possible to further develop salidroside as a new anti-ovarian cancer drug.
