赤麂成纤维细胞的细胞动力学和姐妹染色单体交换率的研究

摘　　要：近几年来,国内外开始应用姐妹染色单体分化(SCD)染色技术研究人和哺乳动物细胞的动力学(Bianchi等1976;Craig-Holmes等1976;Crossen等1977:朱炳富等1980),染色体DNA的复制,包括早复制和晚复制(Lau等1980),细胞周期的测定(等1971;田竟生等1978;等1979),和各种诱变致癌剂的测定(Bloom等1975;Allen等1976;Carrano等1978)取得了许多可喜的结果。本文采用BUdR-吖啶橙-紫外线+Giemsa(BUdR-Acridine Orange-UpG)染色显示姐妹染色单体分化染色技术(贺维顺等1980),初步观察了赤麂肺、皮肤和肾成纤维细胞动力学变化和姐妹染色单体交换率。The cytokinetics and sister chromatid exchange rate of red (Indian) muntjac fibroblasts are studied by the BUdR-UpG (5-bromodeoxyuridine-UV-rays plus Giemsa) method for demonstrating sister cbromatid differential staining.The three kinds of fibroblasts are from lung,skin and kidney tissue of red muntjac.The cytokinetics in lung fibroblasts of red muntjac is observed.During 28-36h,the 1st division metaphases attain 99.5-83.5%,the 2nd division metaphases 1.0-16.5%.During 48-60h,the 1st division metaphases are 69.4-54.1%,the 2nd division metaphases 30.3-41.9%,the 3rd division metaphases 0.3-4.0%.During 72-96h,the 1st division metapnases are 54.0-47.5%,the 3rd division metaphases 14.4-27.9%,the metaphases after the 3rd division 0.7-11.4%.The results in skin fibroblasts are given below.During 28-36h,the 1st division metaphases are 100.0-94.8%.At 36h,the 2nd division metaphases 5.2%.During 48-60h,the 1st division metaphases come to 57.3-32.1%,the 2nd division metaphases 42.7-66.7%.The 3rd division metaphases are 1.0% at 60h.During 7296h,the 1st division metaphases are 24.5-25.5%,the 2nd division metaphases 66.0-54.9%,the 3rd division metaphases 8.2-17.6%,the metaphases after the 3rd division 1.4-7.8%.The cytokinetics in kidney fibroblasts of red muntjac is also observed.The results are:During 28-36h,the 1st division metaphases attain 100.0-95.2%.At 36h,the 2nd division metaphases are 4.8%.During 48-60h,the 1st division metaphases are 69.3-55.4%,the 2nd division metaphases 20.7-41.9%.At 60h,the 3rd division metaphases are 2.7%.During 72-96h,the 1st division metaphases reach 57.2-34.8%,the 2nd division metaphases 47.9-26.8%,the 3rd division metaphases 2.7-26.1%.The metaphases following the 3rd division are 11.6-12.3%.According to the cytokinetic changes of red muntjac three fibroblasts the mitotic cycle are estimated to be:lung fibroblast about 22h,skin fibroblast about 20h,and kidney fibroblast about 25h.Sister chromatid exchange rates of three fibroblasts from red muntjac are also studied.The SCE of lung fibroblast are
