摘  要:目的运用网络药理学方法,比较“除痰解毒方”与“益气除痰方”治疗肺癌的作用机制。方法利用TCMSP数据库筛选出方药活性成分,Gene Cards数据库获取肺癌相关靶点,运用Cytoscape、FunRich软件行GO和KEGG富集分析。结果“除痰解毒方”“益气除痰方”与肺癌交集靶点分别为30个与33个。“除痰解毒方”“益气除痰方”在正常肺组织与肺癌的表达分别为70.42%、69.84%与63.38%、63.49%。两方涉及的生物学过程包括代谢、信号转导、细胞凋亡、免疫反应调控等。KEGG分析提示两方均在TRAIL信号通路有最高表达。结论①两药方靶点蛋白在正常肺组织与肿瘤组织均有亲和性,提示药方并非专一作用于癌细胞。②靶点蛋白在与肺及肺相关脏腑有亲和表达,强调中医论治的系统性与整体性。③靶点蛋白还在脾相关脏腑有亲和表达,预示两方对脾虚证更适用,为肺癌“培土生金”论治提供指导。④靶点蛋白均在SPTAN1基因及TRAIL信号通路有最高相关度,可为靶向药物研究提供新思路。Objective To compare the therapeutic mechanism of Chutan Jiedu Decoction and Yiqi Chutan Decoction for the treatment of lung cancer based on network pharmacology.Methods TCMSP database was used for searching the active components of the herbs of Chutan Jiedu Decoction and Yiqi Chutan Decoction.Gene Cards database were used for obtaining the disease targets related to lung cancer.The enrichment analysis of GO-BP and KEGG pathway was performed by Cytoscape software and Funrich software.Results There were 30 and 33 intersection targets of Chutan Jiedu Decoction with lung cancer and Yiqi Chutan Decoction with lung cancer respectively.The expression rates of Chutan Jiedu Decoction and Yiqi Chutan Decoction in normal lung tissue respectively were 70.42%,69.84%respectively,and those in lung cancer tissue were 63.38%,63.49%respectively.The main biological processes of the two prescriptions involved metabolism,signal transduction,apoptosis,immune response regulation.KEGG pathway enrichment analysis indicated that the two prescriptions had the highest expression rate in TRAIL signaling pathway.Conclusion①The target proteins of the two prescriptions have affinity in normal lung tissue and tumor tissue,indicating that the prescriptions do not specifically act on cancer cells.②The target proteins of the two prescriptions express affinity with lung and lung related viscera,showing the systematic and holistic concept of traditional Chinese medicine treatment.③The target proteins have affinity expression with spleen related viscera,indicating that the two prescriptions are indicated for spleen deficiency syndrome and will provide guidance for the treatment of lung cancer with the therapy of cultivating earth(spleen)to generate metal(lung).④Both of the two prescriptions have the highest expression level in the sptan1 gene and TRAIL signaling pathway,which will provide new ideas for targeted drug research.
