畅脉乐胶囊对MCAO大鼠脑组织保护作用研究及mGluR1、GABA_(B)蛋白表达的影响

摘　　要：目的探讨畅脉乐胶囊对脑缺血再灌注损伤(MCAO)大鼠脑组织的保护机制.方法采用线栓法制备MCAO大鼠模型,随机分为3组,畅脉乐组采用灌胃方式给药,每日1次,连续给药7天;假手术组和模型组给予等剂量的生理盐水.采用mNSS法进行神经功能缺陷评分;TTC染色计算梗死面积和HE染色观察脑损伤情况;采用Real-Time PCR法和Western blot法检测mGluR1和GABA_(B) mRNA蛋白表达情况.结果与模型组相比,畅脉乐组神经功能缺损程度评分显著降低,梗死面积显著减少(P<0.05).畅脉乐组mGluR1mRNA和蛋白表达水平明显降低,GABA_(B) mRNA和蛋白表达水平明显升高(P<0.05).结论畅脉乐胶囊可治疗大鼠脑缺血损伤并调节脑组织mGluR1和GABA_(B)蛋白水平.Objective To investigate the protective mechanism of Changmaile capsule on middle cerebral artery occlusion(MCAO)rat brain tissue.Method The MCAO rat model was established by suture occlusion method.The rats were randomly divided into 3 groups.Changmaile group was given CMLC contents by intragastric administration.once a day,for consecutive 7 days.Sham operation group and model group were given equal volume of normal saline.MNss method was used to score neuro-logical deficits.TTC staining was used to calculate infarct volume and HE staining was used to observe brain injury.The mRNA and protein expressions of mGluR1 and GABA_(B) were detected by real-time PCR and Western blot.Results Compared with model group,the score and infarct volume of Changmaile group were significantly decreased(P<0.05).In addition,the mRNA and protein expression levels of mGluR1 in Changmaile Group were significantly decreased,while the mRNA and protein expression levels of GABA_(B) were significantly increased(P<0.05).Conclusion Changmaile capsule can treat cerebral ischemia injury and regulate the levels of mGluR1 and GABA_(B) protein in the brain of rats.
