摘  要:用L615细胞的双微体(DM)转化A9细胞,得到了转化的A9t3细胞系。A9t3与A9细胞在细胞形态上有显著差异。A9t3细胞含有双微体,具有较高的抗氨甲喋呤能力,二氢叶酸还原酶活性增加,出现了A9细胞所没有的蛋白质电泳带。The gene amplification has been found in number of tumor cell lines and tumor cells. The amplification is related to chromosomal aberrations, including DMs and HSRs. Marinello reported that the shape of double minutes (DM) were usually ring-like. In this study, we reported that the A9 cell line were transformed by DNA from double minute chromosome of L615 cells. The transformed cell line, A9t3, was obtained after selection under condition culture medium. The morphology of A9t3 cells was obviously changed. There are much more double minute chromosomes in A9t3 cells. The resistant ability of A9t3 cells to methotrexate was increased. A new protein band appeared in A9t3 cells, which was not found in A9 cells. According to above results we proposed that the methotrexate resistance in cultured cells tend to generate amplified DHFR and oncogenes on double minute chromosomes. The transformated A9t3 cells have two kinds of protein products of DHFR gene and oncogene.
