摘 要:目的在GB5009.208-2016国家标准方法的基础上建立方便快捷的海鱼罐头中组胺的测定方法。方法称取绞碎均匀的鱼样用5%HClO4溶液浸泡2~3h,用电动振荡器振荡后离心,取上清液用NaOH溶液调节pH值至10~12后加正戊醇振荡提取样液中组胺,离心后取上层正戊醇提取液加1+11 HCl溶液振荡提取正戊醇中组胺,离心后弃去上层正戊醇,取2mL HCl提取液用分光光度法测定组胺含量,从而计算出鱼罐头中组胺含量。结果组胺含量在0~20μg/10mL范围呈良好的线性关系,线性方程Y=0.007275X+0.0211,相关系数γ=0.9999,精密度为2.74%~5.23%,加标回收率为92.2%~96.7%,CFAPA-QC056B-1鱼肉粉中组胺残留量检测质控样品测得值为44mg/100g指定值(452.8±64.9)mg·kg^(-1)。结论建立方便快捷的组胺测定方法,精密度和准确度令人满意,操作步骤少,工作效率高,可同时测定大批量样品中组胺含量。OBJECTIVE To establish a convenient and fast method for the measurement of histamine content in canned marine fish was established based on the national standard method GB5009.208-2016.METHODS The samples were soaked in 40mL 5%perchloric acid solution,and centrifuge the sample after oscillating it with an electric oscillator,take supernatant and adjust pH value to 10~12 with sodium hydroxide solution and add n-amyl alcohol to extract histamine of samples,After centrifugation,take 2mL upper n-amyl alcohol and add 1+11 hydrochloric acid solution to extract histamine from n-amyl alcohol,discard the upper layer n-amyl alcohol after centrifugation.Measure the content of histamine in hydrochloric acid by spectrophotometry and calculate the histamine content in canned fish.RESULTS The content of histamine was linear in the range of 0~20μg/10mL,the correlation coefficient 0.9999,the linear equation Y=0.007275X+0.0211,the precision was 2.74%~5.23%,the standard adding recovery ratio was 92.2%~96.7%.The value of histamine residue in CFAPA-QC056B-1 fish powder sample was 44mg/100g(standard value(452.8±64.9)mg·kg^(-1)).CONCLUSION This histamine measurement method was convenient and quick,the precision and accuracy were satisfactory,with few operation steps and high efficiency,this method could simultaneously measure the content of histamine in large quantities of samples.
