摘 要:以提取液中原花青素与多糖的质量分数为指标,通过对溶剂体积分数、吸涨率、浸泡平衡时间、超声辅助时间等因素的考察确定最佳工艺参数。通过测定刺玫果籽中原花青素、多糖提取物对α-葡萄糖苷酶的抑制率及对羟基自由基的清除率考察其体外活性。结果表明,在最佳提取条件下,刺玫果籽原花青素、多糖的提取率均接近95%,刺玫果籽干浸膏中原花青素、多糖的质量分数分别为145.3、361.3 mg/g。刺玫果籽原花青素、多糖提取物抑制α-葡萄糖苷酶活性的能力均强于阳性对照阿卡波糖,对羟基自由基的清除能力均强于维生素C。柱层析法可同时提取刺玫果籽中的原花青素与多糖,具有提取温度低、提取时间短、节约原料和提取溶剂等优点。Using the quality fraction of procyanidins and polysaccharides in extraction solution as assessment indicators,solvent volume fraction,swelling rate,soaking equilibrium time and ultrasonic assisted time and other factors were evaluated to determine the optimal extracting conditions of proanthocyanidins and polysaccharides from the fruit of Rosa davurica Pall.seeds.The activities of procyanidins and polysaccharides in vitro were evaluated according to their inhibition rate to α-glucosidase and removing rate to hydroxyl radical.The results showed that the extraction rates of procyanidins and polysaccharides in the fruit of Rosa davurica Pall.seeds were close to 95% under the optimum extraction conditions,while their quality fractions were 145.3 and 361.3 mg/g,respectively.The inhibitory activities to α-glucosidase in vitro of procyanidins and polysaccharides were higher than that of acarbose,while their clearance abilities to hydroxyl radicals were higher than that of vitamin C.The method of column chromatography could extract proanthocyanidins and polysaccharides from the fruit of Rosa davurica Pall.seeds simultaneously,with the advantages of low extraction temperature,low extraction time,and saving raw materials and extraction solvents.
