隐丹参酮对肝癌移植瘤小鼠的抑瘤作用及对肠道菌群和外周血淋巴细胞亚型的影响

摘  要:目的研究隐丹参酮对肝癌移植瘤小鼠的肿瘤抑制作用,并探讨其对肝癌移植瘤小鼠肠道菌群以及外周血T淋巴细胞亚型的影响。方法45只Balb/c小鼠按照随机数字表法分为3组,即对照组,模型组和隐丹参酮组,每组15只。模型组和隐丹参酮组小鼠通过腋下注射H22细胞建议肝癌移植瘤模型。待移植瘤直径超过1 cm后,隐丹参酮组小鼠腹腔注射隐丹参酮进行治疗,其余两组小鼠腹腔注射同等剂量0.9%氯化钠溶液。治疗2周后,处死3组小鼠,比较3组小鼠体重、肿瘤重量、肠道屏蔽功能指数、肠道菌群含量、多样性与丰富度,以及外周血T淋巴细胞亚型。结果与对照组相比,模型组小鼠体重下降,但隐丹参酮组小鼠体重显著高于模型组,差异均有统计学意义(P<0.05)。隐丹参酮组小鼠肿瘤重量低于模型组,抑瘤率显著高于模型组,差异均有统计学意义(P<0.05)。与对照组相比,模型组小鼠肠道菌群含量均增加,而隐丹参酮组小鼠肠道菌群含量均低于模型组,差异均有统计学意义(P<0.05)。与对照组相比,模型组小鼠肠道屏蔽功能指数均升高,而肠道内容物菌群多样性与丰富度均降低,差异均有统计学意义(P<0.05);隐丹参酮组小鼠肠道屏蔽功能指数均低于模型组,而肠道内容物菌群多样性与丰富度均高于模型组,差异均有统计学意义(P<0.05)。与对照组相比,模型组小鼠外周血CD4^(+)和CD8^(+)T淋巴细胞比率均上升,而CD4^(+)/CD8^(+)值下降,差异均有统计学意义(P<0.05);隐丹参酮组小鼠外周血外周血CD4^(+)和CD8^(+)T淋巴细胞比率较模型组上升,而CD4^(+)/CD8^(+)值较模型组下降,差异均有统计学意义(P<0.05)。结论隐丹参酮可有效抑制肝癌移植瘤小鼠肿瘤的增殖,并有助于提高肝癌移植瘤小鼠肠道屏蔽功能、肠道菌群多样性和丰度,以及提高其细胞免疫功能。Objective To study the tumor-inhibitory effect of cryptotanshinone in mice with liver cancer transplantation tumor,and to explore its effects on intestinal flora and peripheral blood T lymphocyte subtypes in liver cancer xenograft mice.Methods Forty-five Balb/c mice were randomly divided into 3 groups,namely the control group,the model group and the cryptotanshinone group,with 15 mice in each group.The mice in the model group and cryptotanshinone group were injected with H22 cells under the armpit to suggest a hepatocellular carcinoma xenograft model.When the diameter of the transplanted tumor exceeded 1 cm,the mice in the cryptotanshinone group were treated with intraperitoneal injection of cryptotanshinone,and the mice in the other two groups were intraperitoneally injected with the same dose of 0.9%sodium chloride solution.After 2 weeks of treatment,the three groups of mice were sacrificed,and the body weight,tumor weight,intestinal flora content,diversity and abundance,and peripheral blood T lymphocyte subtypes were compared among the three groups.Results Compared with the control group,the weight of the mice in the model group decreased,but the weight of the mice in the cryptotanshinone group was significantly higher than that in the model group,and the differences were statistically significant(P<0.05).The tumor weight of the cryptotanshinone group was lower than that of the model group,and the tumor inhibition rate was significantly higher than that of the model group,and the differences were statistically significant(P<0.05).Compared with the control group,the content of intestinal flora in the model group was increased,while the content of the intestinal flora in the cryptotanshinone group was lower than that in the model group,and the differences were statistically significant(P<0.05).Compared with the control group,the intestinal shielding function index of the mice in the model group was increased,while the diversity and richness of the intestinal flora were decreased,and the differences were statisti
