摘 要:以柠条花为原料,采用超声微波协同萃取法提取柠条花总黄酮,得率为5.80%;AB-8大孔树脂纯化柠条花总黄酮效率最高,纯化后柠条花总黄酮纯度达到77.09%。利用高效液相色谱-质谱联用技术在纯化柠条花总黄酮中鉴定出18种化合物,包括3种黄酮醇苷元、1种二氢黄酮苷元、3种黄酮苷元、1种花色素类物质以及10种其他类物质。柠条花总黄酮对清除DPPH自由基、ABTS自由基和超氧阴离子自由基的半抑制浓度(IC)分别为5μg/mL,8μg/mL,370μg/mL,柠条花总黄酮浓度为40μg/mL时,DPPH自由基清除率、ABTS清除率和超氧阴离子自由基清除率分别为93.07%,99.05%和10.13%。柠条花总黄酮体外呈现出较强还原力,其体外抗氧化活性与抗坏血酸相近。Caragana Intermedia Kuang et H.C.Fu flowers were used as raw material to obtain total flavonoids by using the ultrasonic-microwave synergistic extraction.The extraction rate was 5.8%;Ab-8 macroporous resin has the highest purification efficiency of total flavonoids from Caragana Intermedia Kuang et H.C.Fu flowers,and the purified total flavonoids reaches 77.09%.The ultra-high liquid chromatography-mass spectrometry technology was used to identify 18 compounds from the purified,total flavonoids of Caragana Intermedia Kuang et H.C.Fu flowers,including 3 flavonol aglycones,1 dihydroflavonoid aglycone,3 flavonoid aglycones,1 anthocyanin,and 10 other types of substances.The ICof total flavonoids of Caragana Intermedia Kuang et H.C.Fu flowers on DPPH,ABTS and superoxide anion free radicals were 5μg/mL,8μg/mL and 370μg/mL,respectively.At 40μg/mL,the radicals scavenging rate of DPPH,ABTS and superoxide anion free were 93.07%,99.05%and 10.13%.It showed a strong reducing power in vitro,and its antioxidant activity in vitro was similar to that of ascorbic acid.
