摘 要:目的 观察人下咽鳞癌FaDu细胞整合素亚单位α(integrin subunitα, ITGA3)表达变化,探讨其对FaDu细胞迁移和侵袭能力的影响。方法 取对数生长期人下咽鳞癌FaDu细胞,分为转染组(转染siRNA-ITGA3质粒)、空载组(转染ITGA3空载质粒)、空白对照组(不作处理);取对数生长期人鼻咽上皮细胞NP69为正常细胞组(不作处理)。转染48 h, 4组采用实时荧光定量PCR法检测ITGA3 mRNA相对表达量,采用Western blot法检测ITGA3蛋白相对表达量;转染组、空载组、空白对照组采用细胞划痕实验检测细胞迁移能力,采用Transwell小室实验检测细胞侵袭能力,采用Western blot法检测基质金属蛋白酶(matrix metalloproteinase, MMP)-2、MMP-9蛋白相对表达量。结果 转染48 h,转染组(0.19±0.04、0.36±0.06)、空载组(0.95±0.13、0.91±0.10)、空白对照组(1.04±0.19、0.95±0.12)ITGA3 mRNA及蛋白相对表达量均高于正常细胞组(0.14±0.02、0.28±0.02)(P<0.05),转染组均低于空白对照组、空载组(P<0.05)。转染48 h,转染组细胞迁移率[(0.27±0.09)%]及MMP-2、MMP-9蛋白相对表达量(0.12±0.03、0.10±0.02)均低于空载组[(0.53±0.06)%、0.42±0.05、0.37±0.05]和空白对照组[(0.55±0.08)%、0.43±0.06、0.36±0.05](P<0.05),侵袭细胞数[(104.73±14.68)个]少于空载组[(210.29±20.14)个]和空白对照组[(214.54±21.56)个](P<0.05),空载组与空白对照组比较差异无统计学意义(P>0.05)。结论 FaDu细胞ITGA3表达上调,下调ITGA3表达可抑制FaDu细胞迁移和侵袭,其机制可能与抑制MMP-2、MMP-9蛋白表达有关。Objective To observe the expression of integrin subunit α(ITGA3) in hypopharyngeal squamous carcinoma FaDu cells, and to investigate its influences on the migration and invasion of FaDu cells. Methods FaDu cells in logarithmic growth phase were divided into transfection group(transfected with siRNA-ITGA3 plasmid), empty group(transfected with ITGA3 empty plasmid) and blank control group(with no treatment). NP69 cells in logarithmic growth phase received no treatment(normal control group). After 48-h transfection, the relative expressions of ITGA3 mRNA and protein were detected by real-time fluorescence quantitative PCR and Western blot, respectively. The cell migration and invasion ability were detected by cell scratch and Transwell assay, and the relative expressions of matrix metalloproteinase(MMP)-2 and MMP-9 were detected by Western blot in transfection group, empty group and blank control group. Results The relative expressions of ITGA3 mRNA and protein were lower in transfection group(0.19±0.04, 0.36±0.06), empty group(0.95±0.13, 0.91±0.10) and blank control group(1.04±0.19, 0.95±0.12) than those in normal control group(0.14±0.02, 0.28±0.02)(P<0.05), and lower in transfection group than those in blank control group and empty group(P<0.05). After 48-h transfection, the cell migration rate, number of invasive cells and relative expressions of MMP-2 and MMP-9 proteins were lower in transfection group [(0.27±0.09)%, 104.73±14.68, 0.12±0.03, 0.10±0.02] than those in empty group [(0.53±0.06)%, 210.29±20.14, 0.42±0.05, 0.37±0.05] and blank control group [(0.55±0.08)%, 214.54±21.56, 0.43±0.06, 0.36±0.05](P<0.05), and showed no significant differences between empty group and blank control group(P>0.05). Conclusion The expression of ITGA3 in FaDu cells is up-regulated, and the down-regulation of ITGA3 expression could supress the migration and invasion, probably by inhibiting the expressions of MMP-2 and MMP-9 proteins.
