摘 要:目的:研究含热蛋白结构域的NOD样受体蛋白1(NLRP1)在头颈部鳞癌(HNSCC)中的表达、预后价值及免疫相关性。方法:通过基于癌症基因组图谱(TCGA)数据库的多个在线网站,分析NLRP1表达及在HNSCC中的预后价值,并通过GO、KEGG分析,明确其参与的生物过程及信号通路。结果:NLRP1在HNSCC中上调,NLRP1的高表达与较长的总生存期呈正相关。功能网络分析及共表达分析表明,NLRP1与先天免疫及炎症等相关,在多个免疫相关的信号通路中发挥作用。此外,在HNSCC中,NLRP1的表达与中性粒细胞、CD4^(+)T细胞、树突状细胞的浸润呈正相关,NLRP1与HNSCC中的多种免疫标志物相关。结论:NLRP1在HNSCC中高表达,NLRP1的高表达可改善预后,可作为HNSCC的潜在预后标志物。NLRP1在先天免疫反应和炎症反应中发挥作用,NLRP1高表达与HNSCC中CD4+T细胞、中粒细胞、树突状细胞的浸润程度呈正相关。PURPOSE:To explore the prognostic value and mechanism of NLR family, pyrin domain containing 1(NLRP1) in head and neck squamous cell carcinoma(HNSCC). METHODS:This study performed bioinformatics analysis on HNSCC data from The Cancer Genome Atlas(TCGA), and jointly analyzed with online databases such as TIMER, TCGA-portal, Kaplan-Meier Plotter, LinkedOmics, GeneMANIA, Metascape, TISIDB and miRDB. RESULTS:NLRP1 was upregulated in HNSCC. Kaplan-Meier survival analysis indicated that patients with high NLRP1 expression had a better prognosis than those with low expression. Furthermore, NLRP1 was concentrated in multiple immune-associated signaling pathways and the expression of NLRP1 was associated with the infiltration of CD4^(+)T cells, neutrophils and dendritic cells.CONCLUSIONS:Our findings confirmed that NLRP1 is highly expressed in HNSCC. The high expression of NLRP1 was significantly related to prognosis and the infiltration degree of CD4+T cells, neutrophils and dendritic cells in HNSCC.
