摘 要:目的:探讨TGF-β1和CD155在舌鳞癌中的表达及相关性,验证TGF-β1是否调控CD155表达,激活ERK1/2通路,对舌鳞癌的生物学行为产生影响。方法:采用免疫组织化学(IHC)方法检测TGF-β1与CD155在46例舌鳞癌中的表达,分析其与临床特征的关系。培养TSCCA、CAL27细胞,分为4组(对照组、TGF-β1组、U0126组、TGF-β1+U0126组),分别采用CCK-8、划痕实验、Transwell实验检测细胞增殖、迁移和侵袭能力,应用Western免疫印迹法检测CD155及P-ERK1/2蛋白表达。采用SPSS 22.0软件包对数据进行统计学分析。结果:TGF-β1和CD155与TNM分期及颈淋巴结转移相关(P<0.05);TSCCA、CAL27细胞中TGF-β1组的增殖能力显著增强(P<0.05),加入通路抑制剂U0126后增殖能力显著减弱(P<0.05);TSCCA、CAL27中TGF-β1组的迁移能力显著增强(P<0.01),U0126组和TGF-β1+U0126组迁移能力显著减弱(P<0.01);TSCCA、CAL27中TGF-β1组的侵袭能力显著增强(P<0.01),U0126组和TGF-β1+U0126组侵袭能力显著减弱(P<0.01);TGF-β1组CD155、 P-ERK1/2蛋白表达显著增加(P<0.05),U0126组和混合干预组CD155、 P-ERK1/2蛋白表达显著较少(P<0.01)。结论:TGF-β1上调CD155表达,激活ERK1/2通路,对舌鳞癌的生物学行为产生影响。PURPOSE:To investigate the expression and relationship of TGF-β1, CD155 in tongue squamous cell carcinoma by histology;to verify cytologically whether TGF-β1 regulates CD155 expression to activate ERK1/2 pathway has an impact on the biological behavior of tongue squamous cell carcinoma. METHODS:IHC was performed to detect the expression of TGF-β1 and CD155 in tongue squamous cell carcinoma and analyze their relationship with clinical data. TSCCA and CAL27 cells were cultured and divided into 4 groups, cell proliferation, migration and invasion of tongue squamous carcinoma cells were detected by CCK-8, scratch and Transwell assays, respectively. Western blot assay was performed to detect CD155and P-ERK1/2 protein expression. SPSS 22.0 software package was used for data analysis. RESULTS:TGF-β1 and CD155were associated with TNM stage and cervical lymph node metastasis(P<0.05). The proliferation ability was significantly enhanced in TSCCA and CAL27 cells in the TGF-β1 group(P<0.05) and weakened by the addition of the pathway inhibitor U0126(P<0.05);the migration ability of TGF-β1 group in TSCCA, CAL27 was significantly enhanced(P<0.01), and weakened in U0126 and TGF-β1+ U0126 groups(P<0.01);the invasion ability of TGF-β1 group was significantly enhanced in TSCCA and CAL27(P<0.01), and weakened in U0126 and TGF-β1+ U0126(P<0.01). Western blot results showed that CD155 and PERK1/2 protein expression was increased in TGF-β1 group(P<0.05), and CD155 and P-ERK1/2 protein expression was less in U0126 group and mixed intervention group(P<0.01). CONCLUSIONS:TGF-β1 up-regulated CD155 expression and activated ERK1/2 pathway has an impact on biological behavior of tongue squamous cell carcinoma.
