摘  要:目的将近红外光谱快速检验方法和BP(Back Propagation)神经网络智能算法相结合,进行长春西汀注射液的NIR-BP联合定量建模方法研究,实现对长春西汀的定量快速检验。方法选取78批长春西汀注射液样品,按照标准方法测定样品含量并采集近红外漫反射光谱。通过优化确定采用多元散射校正MSC光谱预处理方法,建模谱段为6000~8000 cm^(-1),神经网络隐含层神经元20个,隐含层和输出层激励函数分别为purelin和tansig时,所建立的长春西汀注射液的NIR-BP联合定量模型对总体样本预测值与真实值的相关系数R最高。结果所建立的NIR-BP联合定量模型对预测集的预测值与真实值的最大绝对偏差为0.27 mg·mL^(-1),平均绝对偏差为0.10 mg·mL^(-1),最大相对偏差为4.54%,平均相对偏差为1.64%,预测结果与标准检验方法检测结果无显著性差异。对长春西汀注射液和烟酸注射液样本的欧氏距离检验表现出良好的专属性。结论所建立的定量模型均有准确、快速、便捷的特点,提供了近红外光谱建模的新方法,同时对药品的在线检验具有较高的参考价值。Objective To study the near infrared spectrometry-back propagation(NIR-BP)united quantitative analysis modeling of Vinpocetine Injections by using near infrared spectroscopy method combined with back-propagation artificial neural network to achieve the goal of quick online determination.Methods The contents of 78 batches of Vinpocetine Injection samples were obtained by standard inspection method and the near infrared diffuse reflection spectrum of the samples were collected.Optimizations of the parameters combination which can get the highest value of correlation coefficient between the predictive values and the true values were selected for further modeling,which indicates the pretreatment method was the multiplicative scatter correction method,the spectrum range was 6000-8000 cm^(-1),the number of the hidden layer was 20 and the transfer function of the hidden layer and the output layer were purelin and tansig.Results The maximal and average absolute deviations between the predictive values and the true values were 0.27 mg·mL^(-1) and 0.10 mg·mL^(-1),and the relative deviations were 4.54%and 1.64%.The predictive results showed no significant difference with the standard inspection results.Euclidean distance examination of Vinpocetine Injections and Nicotinic Acid Injections showed good specificity.Conclusion The built NIR-BP united quantitative model can determine the contents of the samples precisely,quickly and conveniently,which provides a new method for NIR spectrum quantitative modeling,and supplies a valuable reference for drug online determination.
