婴幼儿配方粉中生物素提取方法与微生物法测定精密度的研究

摘　　要：应用微生物法测定婴幼儿配方乳粉中的生物素,对其样品提取条件进行研究。最终确定在样品中加入50 mL,0.5 mol/L硫酸溶液,在121℃高压灭菌水解30 min后,生物素含量最高。用此提取条件,对6个不同品类的婴幼儿配方粉中生物素进行测定,每个品类样品测定7次,同时由5家实验室用相同方法、相同样品进行实验室间重复性和再现性的验证。最终确定此微生物法测定生物素的精密度为:重复性标准差S_(rj)=28.82%;再现性标准差S_(Rj)=29.80%。The biotin in infant formula was was by by sample extraction conditions.It was finally determined that 50 mL of 0.5 mol/L of sulfuric acid solution was added to the sample,with the highest biotin content after 121℃of autoclaving hydrolysis for 30 min.Using this extraction condition,biotin was measured in 6 different categories of infant formula powder,and each category sample was measured 7 times.Meanwhile,5 laboratories verified the interlaboratory repetition and reproducibility with the same method and the same sample.The final precision of biotin determination by this microbial method is:reproducible standard deviation S_(rj)=28.82%,and reproducibility standard deviation S_(Rj)=29.80%.
