摘  要:目的探讨游泳运动对β淀粉样前体蛋白(APP)/早老素1(PS1)双转基因小鼠闭锁小带蛋白1(ZO-1)、闭合蛋白(occludin)表达和学习记忆的影响。方法将20只APP/PS1小鼠随机分为模型组和游泳组每组10只,对照组选取同窝阴性小鼠10只。游泳组采用游泳运动干预,用新物体识别实验、免疫蛋白印迹法以及免疫荧光检测小鼠学习记忆能力、海马紧密连接蛋白ZO-1、occludin表达和海马CA1区神经元表达情况;硫黄素S染色法检测海马β淀粉样蛋白(Aβ)斑块沉积情况。结果与对照组比较,模型组辨别指数(RI)明显降低(P<0.01);与模型组比较,游泳组RI明显升高[(63.63±13.35)%vs(50.38±9.83)%,P<0.01]。与对照组比较,模型组ZO-1、occludin明显降低(P<0.05,P<0.01);与模型组比较,游泳组ZO-1、occludin明显增加(0.81±0.02 vs 0.38±0.08,1.07±0.03 vs 0.68±0.12,P<0.05)。与对照组比较,模型组神经元阳性表达明显减少(P<0.01);与模型组比较,游泳组神经元阳性表达明显增加,海马Aβ斑块沉积明显减少,差异有统计学意义(P<0.05)。结论游泳运动能够增加APP/PS1小鼠海马紧密连接蛋白ZO-1和occludin蛋白,减轻海马神经元损伤和降低海马Aβ沉积,改善APP/PS1小鼠学习记忆能力。Objective To determine the effects of swimming exercise on the expression of tight junction protein ZO-1 and ocludin and learning and memory abilities in APP/PS1 mouse model of AD.Methods A total of 20 APP/PS1 mice were randomly divided into model and swimming groups,with 10 mice in each group,while another 10 negative mice from the same litter served as the control group.The swimming group was given a swimming exercise intervention.The learning and memory abilities were measured with new object recognition assay,the hippocampal expression levels of ZO-1 and ocludin were detected with Western blotting,the neurons in the hippocampal CA1 region was observed with immunofluorescence assay,and hippocampal Aβplaque deposition was detected after thioflavin S staining.Results The RI was reduced in the model group than the control group(P<0.01),and was increased in the swimming group than the model group[(63.63±13.35)%vs(50.38±9.83)%,P<0.01].Compared with the control group,ZO-1 and ocludin levels were decreased in the model group(P<0.05,P<0.01).The expression of ZO-1 and ocludin was increased in the swimming group than the model group(0.81±0.02 vs 0.38±0.08,1.07±0.03 vs 0.68±0.12,P<0.05).The results of immunofluorescence assay indicated that the number of NeuN in CA1 region of hippocampus was less in the model group than the control group(P<0.01),but larger in the swimming group than the model group(P<0.05).The Aβdeposition was milder in the swimming group than the model group(P<0.05).Conclusion Swimming exercise increases the hippocampal expression of ZO-1 and ocludin proteins in APP/PS1 mice,reduces hippocampal neuronal damage and hippocampal Aβdeposition,and thereby improves the learning and memory abilities in the APP/PS1 mice.
