摘  要:目的 探讨在“手十二井穴”做放血预处理对急性高原低氧肺损伤模型大鼠RAS相关因子表达的影响及作用机制。方法 按照随机数字表法,将72只SD雄性大鼠分为对照组、模型组(6、24、48、72 h组)和放血组(6、24、48、72 h组),每组各8只。采用实验动物低压模拟舱(模拟海拔7 000 m)制备急性高原低氧肺损伤大鼠模型。放血组大鼠按少商-商阳-中冲-关冲-少冲-少泽的顺序,用改良采血针做放血预处理,每日1次,共5次;对照组与模型组除无放血外,其他程序同放血组。用电镜观察大鼠肺组织超微结构;采用ELISA法检测大鼠肺组织血管紧张素Ⅱ(AngⅡ)与内皮素1(ET-1)含量;采用RT-PCR法检测大鼠肺组织血管紧张素转化酶(ACE)mRNA的表达情况。结果 与对照组比较,模型组各时段AngⅡ、ET-1含量和ACE mRNA的表达水平均显著增高,72 h时达高峰。与同时段模型组比较,放血组各时段AngⅡ、ET-1含量下降,ACE mRNA表达水平(72 h组除外)降低。结论 在“手十二井穴”做放血预处理对急性高原低氧肺组织损伤模型大鼠有一定保护作用,以6、24 h为著,其机制可能与下调AngⅡ、ET-1含量及ACE mRNA表达水平有关。Objective To investigate the effect and mechanism of “hand twelve well-points of acupuncture” bloodletting preconditioning on the expression of RAS-related factors in lung tissue injury induced by acute plateau hypoxic in model rats.Methods According to the random number table method,72 SD male rats were divided into control group,model group(6,24,48,72 h)and bloodletting group(6,24,48,72 h),with 8 rats in each group.The experimental animal low-pressure simulation chamber(7 000 m above sea level)was used to prepare a rat model of acute high altitude lung injury.The rats in the bloodletting group were pretreated with a modified blood collection needle in the order of acupuncture points at Shaoshaang-Shangyang-Zhongchong-Guanchong-Shaochong-Shaoze,once a day,for a total of 5 times.Except for no bloodletting,the other procedures were the same as those of the bloodletting group in the control group and the model group.Observed the ultrastructural changes of lung tissue by electron microscope,detected angiotensin Ⅱ(AngⅡ)and endothelin 1(ET-1)contents in rat lung tissue by ELISA,and angiotensin-converting enzyme(ACE)mRNA expression results in rat lung tissue by RT-PCR.Results Compared with the control group,the content of AngⅡ,ET-1 and the expression of ACE mRNA in the model group increased significantly in each time period,and reached the peak at 72 h.Compared with the model group at the same time period,the contents of AngⅡ and ET-1 in the bloodletting group decreased in each period,and the expression of ACE mRNA(except the 72 h group)decreased.Conclusion Bloodletting pretreatment at “hand twelve well-points of acupuncture” has a certain protective effect on lung tissue injury induced by acute plateau hypoxia model rats,and the mechanism is likely to be related to the down-regulation of AngⅡ,ET-1 content and ACE mRNA.
