miR-1262、miR-424表达与上皮性卵巢癌临床病理特征及预后的关系

摘　　要：目的 miR-1262、miR-424表达与上皮性卵巢癌临床病理特征及预后的关系。方法 选择上皮性卵巢癌患者85例。使用问卷调查法调查患者年龄。查阅患者病案信息,收集患者的肿瘤直径、淋巴结转移、FIGO分期、分化程度、组织类型。抽取患者肘静脉血3 ml,离心处理后留下血清,采用Qrt-pcr法检测血清miR-1262、miR-424相对表达量。根据miR-1262、miR-424表达情况,分为低表达和高表达。结果 miR-1262表达与年龄、淋巴结转移无相关性(P>0.05),与FIGO分期、分化程度、组织学类型有关(P<0.05)。miR-424表达与年龄无关(P>0.05),与淋巴结转移、FIGO分期、分化程度、组织学类型有关(P<0.05)。miR-1262低表达患者生存时间(8.02±0.58)个月,高表达患者(6.57±0.51)个月;miR-424低表达患者生存时间(8.14±0.51)个月,高表达患者(6.01±0.58)个月。miR-1262低表达患者生存时间长于高表达者(P<0.05)。miR-424低表达患者生存时间长于高表达者(P<0.05)。miR-1262表达与预后生存时间呈负相关(P<0.05),miR-424表达与预后生存时间呈正相关(P<0.05)。结论 上皮性卵巢癌患者miR-1262、miR-424高表达,其表达水平与疾病临床病理特征及预后密切。miR-1262、miR-424高表达会促使肿瘤发生和发展,建议密切灌注。Objective To investigate the correlation of miR-1262 and miR-424 expressions with the clinicopathological characteristics and prognosis in patients with epithelial ovarian cancer.Methods 85 patients with epithelial ovarian cancer were enrolled.Clinical data including age,tumor diameter,lymph node metastasis,FIGO stage,degree of differentiation and tissue type were collected using questionnaire.Blood samples of 3 ml of elbow venous blood were collected,and the relative expression of serum miR-1262 and miR-424 was detected by Real-time reverse transcription PCR(qRT-PCR)after centrifugation.Patients were classified as low and high expression groups according to the detection results.Results The miR-1262 expression had no correlation with age and lymph node metastasis(P>0.05),while had correlation with FIGO stage,degree of differentiation,and tissue type(P0.05),while had correlation with lymph node metastasis status,FIGO stage,degree of differentiation,and tissue type(P<0.05).Survival time was(8.02±0.58)months in patients with low miR-1262 expression,which was significantly longer than(6.57±0.51)months in patients with high miR-424 expression.Survival time was(8.14±0.51)months for patients with low miR-424 expression,which was significantly longer(6.01±0.58)months for patients with high miR-424 expression.miR-1262 expression was negatively correlated with prognostic survival time(P<0.05)and miR-424 expression was positively correlated with prognostic survival time(P<0.05).Conclusion Patients with epithelial ovarian cancer have elevated expression of miR-1262 and miR-424,and their expression levels are closely related to the clinicopathological features and prognosis of the disease.Moreover,high expressions of miR-1262 and miR-424 may promote the tumorigenesis and progression,and so the detection of the two is of great clinical value.
