摘  要:目的 探讨lncRNA TSPOAP1-AS1对缺糖缺氧(oxygen-glucose deprivation, OGD)处理的脑微血管内皮细胞(brain microvascular endothelial cells, BMECs)存活的影响。方法 收集10例缺血性脑卒中(ischemic stroke, IS)患者血清样本和10名同期进行健康体检者血清样本,实时荧光定量PCR(real-time fluorescence quantitative PCR,RT-qPCR)检测样本中TSPOAP1-AS1的表达水平;小鼠BMECs进行正常培养和OGD诱导,RT-PCR检测2组BMECs中TSPOAP1-AS1相对表达水平;干扰OGD诱导的BMECs中TSPOAP1-AS1表达(si-TSPOAP1-AS1+OGD组),对照组为si-NC+OGD组。RT-qPCR检测细胞中TSPOAP1-AS1相对表达水平,MTS法检测细胞培养24、48、72 h后细胞的存活率。结果 IS患者血清TSPOAP1-AS1表达水平显著高于健康体检者(P<0.01);OGD组细胞中TSPOAP1-AS1表达水平显著高于正常培养组(P<0.01);转染si-TSPOAP1-AS1能够显著下调OGD处理后的BMECs中TSPOAP1-AS1相对表达水平(P<0.001)。与si-NC+OGD组相比,si-TSPOAP1-AS1+OGD组细胞在培养24、48、72 h后的存活率均增大(均P<0.01)。结论 干扰TSPOAP1-AS1表达可促进OGD处理BMECs的存活。Objective To investigate the effect of lncRNA TSPOAP1-AS1 on the survival of brain microvascular endothelial cells(BMECs) treated with oxygen-glucose deprivation(OGD). Methods The serum samples from 10 patients with ischemic stroke(IS) and 10 healthy subjects were collected. The expression level of TSPOAP1-AS1 in these samples was analyzed by real-time fluorescence quantitative PCR(RT-qPCR). The mouse BMECs were divided into a normal culture group and an OGD treatment group. The expression level of TSPOAP1-AS1 in the 2 groups was detected by RT-qPCR. Interfering TSPOAP1-AS1 expression in OGD-induced BMECs(si-TSPOAP1-AS1+OGD group), and si-NC+OGD group was the control group, then the relative expression level of TSPOAP1-AS1 in BMECs was characterized by RT-qPCR. The survival rates of BMECs for 24, 48, and 72 h were assessed by MTS assays. Results The expression level of serum TSPOAP1-AS1 in IS patients was significantly higher than that in healthy subjects(P<0.01). The expression level of TSPOAP1-AS1 in OGD group was significantly higher than that in normal culture group(P<0.01). Transfection of si-TSPOAP1-AS1 could significantly down regulated the relative expression level of TSPOAP1-AS1 in BMECs treated with OGD(P<0.001). Compared with si-NC + OGD group, the survival rate of cells in si-TSPOAP1-AS1 + OGD group was significantly increased at 24, 48, and 72 h(P<0.01). Conclusion Interfering TSPOAP1-AS1 expression promotes the survival of OGD-treated BMECs.
