摘  要:目的:探索去氢延胡索素(DHC)对膀胱癌细胞增殖、迁移、侵袭和凋亡的作用机制。方法:通过CCK-8和EdU实验验证DHC对细胞增殖的影响;划痕实验和Transwell迁移和侵袭实验验证DHC对膀胱癌细胞迁移和侵袭的影响;Western blot和Annexin V-FITC/PI双染实验探究DHC对细胞凋亡的影响。结果:与对照组0μmol/L相比,25、50、100和200μmol/L组的DHC可以抑制细胞的活性(P<0.01),增加BAX蛋白的表达(P<0.01),抑制BCL-2蛋白的表达(P<0.01);与对照组相比,DHC可以抑制膀胱癌细胞的增殖、迁移、侵袭并诱导其凋亡(P<0.01)。结论:DHC可以抑制膀胱癌细胞的增殖、迁移、侵袭并诱导其凋亡,提示其可能成为潜在的抗膀胱癌候选药物。Objective:To explore the mechanism of dehydrocorydaline(DHC)in proliferation,migration,invasion and apoptosis of bladder cancer(BCa)cells.Methods:The effect of DHC on cell proliferation was verified by CCK-8 and EdU assays,and on BCa cell migration and invasion was determined by wound healing,transwell migration and invasion assays.Western blot and Annexin V-FITC/PI double staining were applied to observing the effect of DHC on cell apoptosis.Results:Compared with 0μmol/L dose control,DHC in dose of 25,50,100 and 200μmol/L led to inhibited cell activity(P<0.01),up-regulated protein BAX expression(P<0.01),yet inhibited expression of BCL-2 protein(P<0.01).Generally,DHC resulted in inhibition of proliferation,migration,invasion and induction of apoptosis of BCa cells(P<0.01).Conclusion:DHC could inhibit the proliferation,migration,invasion and induce the apoptosis of BCa cells,suggesting that it may be a potential anti-bladder cancer drug candidate.
