摘 要:目的制备得到一种既具有良好温敏效果,又具有自显影特性的聚(N-异丙基丙烯酰胺)-共聚-四碘磺酸水凝胶,可实现对肿瘤血管持续栓塞,其内部的显影功能又有助于影像学的术后随访。方法将四碘-2-磺酸苯甲酸酐与甲基丙烯酸羟乙酯的化合物,接枝在聚(N-异丙基丙烯酰胺)链上,制备得到聚(N-异丙基丙烯酰胺)-共聚-四碘磺酸水凝胶。采用傅里叶变换红外光谱仪、电感耦合等离子体质谱仪、凝胶渗透色谱、扫描电镜、透射电镜对其结构、碘含量、分子量和形貌进行表征,采用动态光散射粒度仪、高速旋转流变仪、UV/Vis分光光度计对其温敏特性进行表征,通过小鼠皮下注射凝胶,来评估材料的生物相容性,最后,采用正常兔肾动脉栓塞模型来评价聚(N-异丙基丙烯酰胺)-共聚-四碘磺酸水凝胶在体内的栓塞效果。结果合成的聚(N-异丙基丙烯酰胺)-共聚-四碘磺酸水凝胶,其溶-凝胶相转变温度为34℃左右,生物相容性良好,兔肾动脉模型显示血管栓塞效果良好。结论聚(N-异丙基丙烯酰胺)-共聚-四碘磺酸水凝胶作为一种液体栓塞材料,可以注入动脉血管并原位凝固以达到栓塞效果,其内在的自显影功能也为影像学的随访观察提供了便捷。这种具有温度敏感性和自显影效果的双功能化水凝胶,有望成为一种新型的医用栓塞剂运用于临床动脉栓塞治疗之中,具有较好的研究价值和应用前景。Objective To prepare a poly(N-isopropylacrylamide)-co-tetraiodosulfonic acid hydrogel with good thermosensitive effect and auto-imaging properties,which can achieve continuous embolization of tumor blood vessels.The internal imaging function is also helpful for postoperative follow-up of imaging.Methods The compound of tetraiodo-2-sulfonic acid benzoic anhydride and hydroxyethyl methacrylate was grafted on the poly[(N-isopropylacrylamide)chain to prepare poly(N-isopropylacrylamide)]-copolymer-tetraiodosulfonic acid hydrogel.Its structure,iodine content,molecular weight and morphology were characterized by Fourier transform infrared spectroscopy,inductively coupled plasma mass spectrometry,gel permeation chromatography,scanning electron microscopy and transmission electron microscopy.Then,using a dynamic light scattering particle size analyzer,a high-speed rotational rheometer,and a UV/Vis spectrophotometer to characterize its temperature-sensitive properties,and subcutaneously injecting the hrdrogel to evaluate the biocompatibility of the material.Finally,a normal rabbit renal artery embolization model was used to evaluate the embolization effect of poly(N-isopropylacrylamide)-copolymer-tetriodosulfonic acid hydrogel in vivo.Results The synthesized poly(N-isopropylacrylamide)-co-tetraiodosulfonic acid hydrogel has a solution-gel phase transition temperature of about 34℃and good biocompatibility.The rabbit renal artery model showed good vascular embolization effects.Conclusion Poly(N-isopropylacrylamide)-co-tetraiodosulfonic acid hydrogel,as a liquid embolization material,can be injected into arterial vessels and coagulated in situ to achieve embolization effect.It provides convenience for follow-up observation of imaging.This bifunctional hydrogel with temperature sensitivity and auto-imaging effect is expected to become a new type of medical embolizing agent used in clinical arterial embolization treatment,and has good research value and application prospect.
