摘  要:β-葡萄糖苷酶在纤维素资源开发、食品加工和糖苷类化合物的合成等方面具有多种用途,为了高效制备该酶,本文以植物种籽为原料,筛选出苹果籽为最佳酶源,在此基础上采用三相法提取其中的β-葡萄糖苷酶,考察了影响三相提取的主要因素。结果表明以正戊醇为有机溶剂,其与粗酶液的体积比为2:1,硫酸氨的饱和度为70%,提取体系的pH值为5.0,提取温度为35℃,最终纯化倍数可达7.51,酶活回收率为88.68%,本研究为规模化制备β-葡萄糖苷酶提供了新方法。β-glucosidase has many uses in the cellulose resource development,food processing and the glycosides synthesis.In order to prepare the enzyme efficiently,the paper uses plant seeds as raw materials and selects apple seeds as the best enzyme source.On this basis,it was extracted by three-phase partitioning.The main factors affecting the three-phase partitioning were investigated.The results showed that the volume ratio of n-pentanol to crude enzyme solution was 2:1,the saturation of ammonia sulfate was 70%,the pH value of the extraction system was 5.0,the extraction temperature was 35℃.Under the optimal conditions,the final purification factor was 7.51 and the enzyme activity recovery was 88.68%This study provides a new method for large-scale preparation ofβ-glucosidase.
