摘  要:为了获得金莲花基因数据,本研究利用Illumina Novaseq 6000高通量测序平台对金莲花叶片进行转录组测序,经de novo组装后共获得了36101条Unigenes,平均长度为1280 bp,N50为1789 bp。进一步利用7个公共数据库(Nr,Nt,Pfam,KOG,Swiss-prot,KEGG,GO)进行比对,分别注释到了21354、13806、16161、5435、16437、36101及16160条Unigenes。注释结果显示,金莲花与洛矶山耧斗菜的同源序列最多;在GO和KOG数据库中,可将其分为3大类42小类和3大类25小类;在KEGG数据库中,36101条Unigenes参与了34类代谢途径,并挖掘出与黄酮合成相关结构基因。对编码序列和微卫星进行检测分析,共发现了16535条CDS和10761个SSR位点。本研究通过转录组测序,获得了大量的基因序列,为进一步挖掘功能基因、开发分子生物学标记和金莲花黄酮类化合物合成途径研究等提供了一定的科学依据。In order to obtain the gene data of Trollius chinensis,the transcriptome of Trollius chinensis leaves was sequenced by using Illumina Novaseq 6000,and 36101 Unigenes were obtained after assembly by de novo,with an average length of 1280 bp and an N50 of 1789 bp.Furthermore,seven public databases(Nr,Nt,Pfam,KOG,Swiss-prot,KEGG,GO)were used for comparison,and 21354,13806,16161,5435,16437,36101 and 16160Unigenes were annotated respectively.The annotation results showed that Trollius chinensis had the most homologous sequences with Aquilegia coerulea.In GO and KOG databases,they can be divided into 3 categories,42 subclasses and 3 categories,25 subclasses.In KEGG database,36101 Unigenes participated in 34 metabolic pathways,and structural genes related to flavone synthesis were excavated.A total of 16535 CDS and 10761 SSR loci were found by detecting and analyzing the coding sequence and microsatellite.In this study,a large number of gene sequences were obtained by transcriptome sequencing,which provided a certain scientific basis for further mining functional genes,developing molecular biological markers and studying the synthetic pathway of flavonoids in Trollius chinensis.
