摘　　要：目的:观察LncRNA MALAT1对Saos-2细胞自噬、增殖、迁移及侵袭的影响,并探讨其作用机制。方法:干扰LncRNA MALAT1在Saos-2细胞中的表达水平,CCK8法检测细胞增殖能力,流式细胞技术检测细胞凋亡及周期情况,细胞划痕实验及Transwell实验检测细胞迁移及侵袭改变,透射电镜观察自噬小体,Western Blot、RT-PCR检测自噬相关因子Beclin1、LC3、p62表达差异及LncRNA MALAT1对Wnt/β-catenin通路的影响。结果:干扰LncRNA MALAT1后,Saos-2细胞自噬水平明显升高,Beclin1、LC3表达水平明显升高(P<0.05),而p62的表达水平显著下降(P<0.05);细胞增殖、迁移和侵袭能力显著下降(P<0.05),Wnt/β-catenin通路相关因子Wnt3a、β-catenin的表达水平显著下降(P<0.05)。结论:LncRNA MALAT1可以通过激活Wnt/β-catenin通路调控Saos-2细胞自噬、增殖、迁移和侵袭。Objective:To observe the effects of LncRNA MALAT1 on autophagy,proliferation,migration and invasion of Saos-2 cells,and to explore its mechanism.Methods:The expression level of LncRNA MALAT1 in Saos-2 cells was interfered,the cell proliferation level was detected by CCK8 method,the apoptosis and cycle were detected by flow cytometry,the changes of cell migration and invasion were detected by cell scratch test and Transwell test,the autophagy bodies were observed by transmission electron microscope,and the autophagy related factors Beclin1,LC3,p62 and Wnt/β-catenin pathway related proteins were detected by Western blot and RT-PCR.Results:After interfered,the autophagy level of Saos-2 cells increased,the expression level of Beclin1 and LC-3 increased significantly(P<0.05),while the expression level of p62 decreased significantly(P<0.05).At the same time,the level of cell proliferation and migration decreased significantly(P<0.05).Furthermore,the expression level of Wnt/β-catenin pathway related factors Wnt3a andβ-catenin decreased significantly(P<0.05).Conclusion:LncRNA MALAT1 can regulate the autophagy,proliferation,migration and invasion of Saos-2 cells via activating Wnt/β-catenin pathway.
