摘　　要：基底样乳腺癌(basal-like breast cancer,BLBC)是乳腺恶性肿瘤中最具侵袭性的分子亚型,好发于年轻女性,与其他亚型相比具有更高的局部和远处转移倾向,多预后不良。临床上约60%~90%的BLBC病理表现为三阴性表型,这意味着大多数患者不适合内分泌治疗或HER2靶向治疗。因此,BLBC的治疗选择有限,迫切需要开发新的预后标志物和治疗靶点。叉头盒蛋白C1(forkhead box C1,FOXC1)是一种参与胚胎发育的重要转录因子。近年来研究发现,FOXC1在多种肿瘤中高表达并发挥癌基因的作用。在乳腺癌中,FOXC1特征性的在BLBC亚型中表达升高,并与BLBC的侵袭性生物学行为和远处转移倾向密切相关,是导致BLBC预后较差的重要因素。深入研究发现,FOXC1通过复杂的机制调控网络影响BLBC的发生发展和对治疗的反应,对FOXC1的干预有望成为诊疗BLBC的新方向。然而,目前对FOXC1的作用机制研究仍处于初级阶段。因此,本文就近年来发表的相关研究,对FOXC1在BLBC中的功能及临床应用的进展作一综述,旨在为将来研究方向的选择提供参考。Basal-like breast cancer(BLBC),as the most malignant subtype of breast cancer,is more commonly diagnosed in young women,and has a higher propensity for local and distant metastases compared with other subtypes,resulting in a poor prognosis.About 60%to 90%of BLBC patients demonstrate triple-negative features,which means that most patients are resistant to endocrine therapy or HER2-targeted therapy.Therefore,the treatment options for BLBC are limited.It is urgent to develop new prognostic markers and therapeutic targets.Forkhead box protein C1(FOXC1)is known to relate to embryonic development.Recent studies have found that it is highly expressed in various tumors and acts as an oncogene.In breast cancer,FOXC1 is specifically elevated in BLBC subtypes,which associates with aggressive biological behavior and propensity of metastasis,indicating a poor prognosis.In-depth studies have found that FOXC1 affects the occurrence and development of BLBC and the response to treatment through a complex regulatory network.The intervention of FOXC1 is expected to become a new direction for the diagnosis and treatment of BLBC.However,the specific mechanism of FOXC1 in BLBC is still in the developmental stage.Therefore,this review summarizes function and current clinical application of FOXC1 in BLBC,in order to provide a reference for the selection of future research directions.
