摘 要:目的探讨亲环蛋白D(cyclophilin D,CypD)在甲基乙二醛(methylglyoxal,MG)诱导成骨细胞凋亡中的作用及机制。方法本研究将细胞随机分为对照组、MG组、环孢菌素A(CsA)组和CsA+MG组。采用MTT法检测细胞增殖活性,TUNEL染色法检测细胞凋亡,Western blot法检测CypD蛋白表达水平,TMRM染色法检测线粒体膜电位。结果MTT结果表明,MG抑制了成骨细胞增殖活性,并与浓度呈正相关。TUNEL染色结果显示,MG组较对照组显著增加了成骨细胞凋亡,而Western blot法检测结果表明,MG处理后CypD蛋白表达增加。在加入CypD抑制剂CsA干预后,其显著恢复了MG抑制的细胞增殖活性,细胞凋亡减少,CypD蛋白表达水平降低。同时,与MG组比较,TMRM结果表明,CsA部分恢复线粒体膜电位强度。结论MG可能通过调控CypD蛋白来诱导成骨细胞凋亡。Objective To explore the effect and mechanism of methylglyoxal(MG)on osteoblast apoptosis by regulating cyclophilin D(CypD)expression.Methods Logarithmic growth phase MC3T3-E1 were divided into 4groups:control group,MG group,cyclosporin A(CsA)group,CsA+MG group.Cell viability was evaluated by MTT assays.The apoptosis rate was analyzed by TUNEL assays.The protein expression level of CypD was detected by Western blot.TMRM staining was used to evaluate the mitochondrial membrane potential.Results As compared with control group,MG decreased the number of viable cells.TUNEL staining showed an increase in the incidence of apoptosis.What′s more,MG increased the expression of CypD.As expected,CsA pretreatment greatly improved the viability of cells exposed to MG and attenuated MG-induced apoptosis.Furthermore,CsA treatment markedly decreased level of CypD and raised mitochondrial membrane potential.Conclusion MG induced osteoblast apoptosis,possibly by regulating CypD expression.
