基于lncRNA BANCR介导MMPs的表达探讨夏枯草对甲状腺乳头状癌增殖、侵袭和迁移的影响

摘　　要：目的研究长链非编码RNA BANCR及基质金属蛋白酶MMPs在甲状腺乳头状癌(papillary thyroid carcinoma,PTC)中的表达,进而探讨探究夏枯草对PTC细胞增殖、迁移和侵袭的影响。方法培养甲状腺乳头状癌细胞和正常甲状腺细胞,lncRNA BANCR siRNA和si-NC分别转染细胞进行分组。qRT-PCR技术检测lncRNA BANCR的表达水平,Western blot法检测MMP2、MMP9表达水平;CCK-8、Transwell以及划痕实验用以检测夏枯草对B-CPAP细胞增殖、侵袭与迁移的影响。结果B-CPAP细胞相较于正常甲状腺细胞,lncRNA BACNR和MMP2、MMP9表达上调(P<0.05),中药组与对照组比较,lncRNA BACNR和MMP2、MMP9表达水平下降(P<0.05),B-CPAP细胞的增殖、侵袭和迁移能力受到抑制(P<0.05)。结论中药夏枯草能够通过下调lncRNA BANCR降低MMP2、MMP9的表达来抑制PTC细胞的增殖、侵袭和迁移。Objective To study the expression of long non-coding RNA BANCR and matrix metalloproteinases MMPs in papillary thyroid carcinoma(PTC),and to explore the effects of Prunella vulgaris on the proliferation,migration and invasion of PTC cells.Methods Cultured papillary thyroid cancer cells and normal thyroid cells,lncRNA BANCR siRNA and si-NC were transfected into the cells to group.The expression level of lncRNA BANCR was detected by qRT-PCR technology.The expression level of MMP2 and MMP9 was detected by Western blot technology.CCK-8,Transwell and scratch experiments were used to detect the effects of Prunella vulgaris on the proliferation,invasion and migration of B-CPAP cells.Results Compared with normal thyroid cells,the expressions of lncRNA BACNR and MMP2 and MMP9 were up-regulated in B-CPAP cells(P<0.05).Compared with the control group,the expression levels of lncRNA BACNR,MMP2 and MMP9 were decreased in the traditional Chinese medicine group(P<0.05).The proliferation,invasion and migration abilities of B-CPAP cells were inhibited(P<0.05).Conclusion Prunella vulgaris can inhibit the proliferation,invasion and migration of PTC cells by down-regulating lncRNA BANCR and reducing the expression of MMP2 and MMP9.
