摘  要:目的研究水飞蓟素(SM)通过Slit2调控非小细胞肺癌(NSCLC)细胞增殖及凋亡的作用机制。方法培养NSCLC细胞株A549,分为对照组及不同剂量SM组(1、20、40 mg/L),转染阴性对照(NC)siRNA的si-NC组、转染NC siRNA及40 mg/L SM干预的si-NC+40 mg/L SM组、转染Slit2 siRNA及40 mg/L SM干预的si-Slit2+40 mg/L SM组。干预48 h后检测细胞增殖A_(490)水平、细胞凋亡率及Slit2、ROBO1、C-X-C基序趋化因子受体4(CXCR4)的蛋白表达水平。结果10、20、40 mg/L SM组A549细胞的A_(490)水平低于对照组,凋亡率高于对照组,差异有统计学意义(P<0.05)。20 mg/L SM组A549细胞的A_(490)水平低于10 mg/L SM组,凋亡率高于10 mg/L SM组,差异有统计学意义(P<0.05)。40 mg/L SM组A549细胞的A_(490)水平低于10、20 mg/L SM组,凋亡率高于10、20 mg/L SM组,差异有统计学意义(P<0.05)。与对照组比较,10、20、40 mg/L SM组A549细胞中Slit2、ROBO1的表达水平升高,CXCR4的表达水平降低,差异有统计学意义(P<0.05)。与10 mg/L SM组比较,20 mg/L SM组A549细胞中CXCR4的表达水平降低,差异有统计学意义(P<0.05)。40 mg/L SM组A549细胞中Slit2、ROBO1的表达水平升高,CXCR4的表达水平降低,差异有统计学意义(P<0.05)。与20 mg/L SM组比较,40 mg/L SM组A549细胞中Slit2、ROBO1的表达水平升高,CXCR4的表达水平降低,差异有统计学意义(P<0.05)。与si-NC组比较,si-NC+40 mg/L SM组A549细胞中Slit2的表达水平及凋亡率升高,A_(490)水平降低,差异有统计学意义(P<0.05);与si-NC+40 mg/L SM组比较,si-Slit2+40 mg/L SM组A549细胞中Slit2的表达水平及凋亡率降低,A_(490)水平升高,差异有统计学意义(P<0.05)。与si-NC组比较,si-NC+40 mg/L SM组A549细胞中ROBO1的表达水平升高,CXCR4的表达水平降低,差异有统计学意义(P<0.05);与si-NC+40 mg/L SM组比较,si-Slit2+40 mg/L SM组A549细胞中ROBO1的表达水平降低,CXCR4的表达水平升高,差异有统计学意义(P<0.05)。结论SM通过激活Slit2/ROBO1通路抑Objective To study the effect and mechanism of silymarin(SM)on proliferation and apoptosis of non-small cell lung cancer(NSCLC)cells through Slit2.Methods NSCLC cell line A549 was cultured and divided into control group and SM groups with different doses(10,20 and 40 mg/L),si-negative control(NC)group transfected with NC siRNA,si-NC+40 mg/L SM group transfected with NC siRNA and intervened with 40 mg/L SM,si-Slit2+40 mg/L SM group transfected with Slit2 siRNA.After 48 hours of intervention,cell proliferation A_(490) level,apoptosis rate,and the expression levels of Slit2,ROBO1,and C-X-C motif chemokine receptor 4(CXCR4)were detected.Results The A_(490) level and apoptosis rate of A549 cells in 10,20 and 40 mg/L SM groups were lower than those in control group,and the differences were statistically significant(P<0.05).The A_(490) level of A549 cells in 20 mg/L SM group was lower than that in 10 mg/L SM group,and the apoptosis rate was higher than that in 10 mg/L SM group,the differences were statistically significant(P<0.05).The A_(490) level of A549 cells in 40 mg/L SM group was lower than that in 10 and 20 mg/L SM groups,and the apoptosis rate was higher than that in 10 and 20 mg/L SM groups,and the differences were statistically significant(P<0.05).Compared with the control group,the expression levels of Slit2 and ROBO1 in A549 cells of 10,20 and 40 mg/L SM groups were increased,and the expression level of CXCR4 was decreased,and the differences were statistically significant(P<0.05).Compared with 10 mg/L SM group,the expression level of CXCR4 in A549 cells in 20 mg/L SM group was decreased,and the difference was statistically significant(P<0.05).The expression levels of Slit2 and ROBO1 in A549 cells in 40 mg/L SM group were increased,and the expression level of CXCR4 was decreased,the differences were statistically significant(P<0.05).Compared with 20 mg/L SM group,the expression levels of Slit2 and ROBO1 in A549 cells in 40 mg/L SM group were increased,and the expression level of CXCR4 was decreased,and the d
