摘 要:膜片钳记录技术是神经电生理研究中的核心技术,是基础科研领域在单细胞水平检测可兴奋细胞(神经元和心肌细胞等)放电活动的重要技术手段。此外,由于膜片钳技术在研究细胞水平生理功能中的权威性和精确性,医学本科生对这一技术的学习和掌握将极大地促进对“细胞的基本功能”这一章节相关内容的理解和应用,并为后续的理论学习打下良好的基础。目前,受限于较高的技术硬件要求及实验操作门槛,基于该技术的实验教学在本科学生中普遍开展仍具有较大难度。陆军军医大学(以下简称“我校”)生理教研室在长期成熟应用该技术的基础上,首先在我校生物技术专业本科学生中尝试开展了膜片钳技术实验教学,并于近年将其广泛应用到医学本科生生理创新实验班的教学中。实践教学中,我校生理教研室力求在传统“验证性”生理教学实验中突出对学生兴趣和创新实践能力的引导。本文将近年来的教学方式、内容及经验进行总结探讨,力求为下一步推广该实验教学提供借鉴和基础。Patch-clamp recording technology is the core technology in neuroelectrophysiological research,and it is an important technical means to detect the firing activity of excitable cells(neurons and cardiomyocytes,etc.)at the single cell level in the field of basic scientific research.In addition,due to the authority and accuracy of patch-clamp technology in the study of physiological functions at the cell level,the study and mastery of this technology for medical undergraduates will greatly promote the understanding and application of the chapter“basic functions of cells”,and lay a good foundation for the subsequent theoretical study.Currently,due to the high hardware requirement and complex operation procedures,it is still very difficult to apply this technique in the experimental course of medical undergraduate students.The department of physiology in the Army Medical University(hereinafter referred to as“our school”)has applied patch-clamp recording in scientific research for a long time.On this basis,the department of physiology of our school has made efforts to apply this important technique in the experimental course of undergraduate students who major in biotechnology first,and then apply it in the physiological innovation experimental class of medical undergraduate students in recent years.In this process,the department of physiology of our school aim at arousing the students’interest for basic research and cultivating their innovative practice ability.Here,the present papers have summarized the contents,methods,as well as experiences during this teaching exploration,in order to further improve the introduction of patch-clamp recording technique into the experimental course of undergraduate students.
