摘  要:目的通过观察刺梨冻干粉对慢性肾衰竭模型大鼠微炎症状态的影响,探讨刺梨对肾脏的保护机制,是否和调节JAK/STAT信号通路,使慢性肾衰竭模型大鼠微炎症状态得到有效改善相关。方法使用腺嘌呤灌胃法造模,大鼠依据体重分层,随机分为正常组和造模组。造模后第2 d,将造模组大鼠随机分为刺梨组、模型组及氯沙坦钾组,全部大鼠参考马氏定理设置干预剂量,氯沙坦钾组给药10mg/(kg·d),刺梨组给药3 g/(kg·d),浓度为3%。模型组和正常组均同时给予等量的无菌生理盐水,连续14 d。ELISA法检测各组IL-6、p-JAK2、p-STAT3水平浓度。结果与正常组比较,造模组大鼠目标指标p-JAK2、p-STAT3、IL-6浓度均明显升高(P<0.01);与模型组比较,刺梨组、氯沙坦钾组上述各项指标均明显降低(P<0.01);与刺梨组比较,氯沙坦钾组对指标数值的降低作用更明显(P<0.01)。结论氯沙坦钾和刺梨可能通过抑制p-JAK2/p-STAT3信号通路的激活减少IL-6等炎症因子的表达,调节免疫微环境,减轻CRF模型大鼠微炎症状态,从而起到一定的肾脏保护作用。Objective:To study the protective mechanism of Rosa Roxbunghii on the kidney by observing the effect of freeze-dried powder of Rosa Roxbunghii on the micro-inflammatory state of chronic renal failure model rats,and whether it regulates the JAK/STAT signaling pathway,so that the micro-inflammation of chronic renal failure model rats can be effectively improved.Methods:Models were established by adenine gavage.Rats were stratified according to body weight and randomly divided into normal group and modeling group.On the 2nd day after modeling,the rats in the modeling group were randomly divided into a Rosa group,a model group and a losartan potassium group.(kg·d).All rats were given the intervention dose according to Markov's theorem.The losartan potassium group was administered 10 mg/(kg·d).The Rosa group was given 3 g/(kg·d)and the concentration was 3%.The model group and the normal group were given the same amount of sterile normal saline at the same time for 14 consecutive days.The levels of IL-6,p-JAK2 and p-STAT3 in each group were detected by ELISA.Results:Compared with that of the normal group,the concentrations of target indexes p-JAK2,p-STAT3 and IL-6 in the model group were significantly increased(P<0.01);compared with that of the model group,all the above indexes of the Rosa group and the losartan potassium group were significantly decreased(P<0.01);compared with the Rosa group,the losartan potassium group had a more significant decrease in the index values(P<0.01).Conclusion:Losartan potassium and Rosa roxbunghii may reduce the expression of IL-6 and other inflammatory factors by inhibiting the activation of p-JAK2/p-STAT3 signaling pathway,regulate the immune microenvironment,and reduce the micro-inflammatory state of CRF model rats,thereby playing a role in reducing the expression of IL-6 and other inflammatory factors,with a certain kidney protection.
