摘  要:目的:优选胶体磨辅助酶提取鹿茸可溶性蛋白的最佳工艺并验证提取物的免疫活性。方法:以鹿茸蛋白提取率为指标,通过单因素试验研究胶体磨磨齿间隙、胶体磨研磨时间、加酶量、酶解温度、酶解时间等因素对蛋白提取率的影响,在此基础上利用正交试验设计,优化提取工艺条件,并通过免疫活性试验确定其免疫活性。结果:胶体磨辅助酶提取马鹿茸可溶性蛋白的最佳提取工艺为胶体磨磨齿间隙为50μm,胶体磨研磨时间30min,加酶量为40mg/g,酶解时间40min,酶解温度50℃;体内免疫活性试验表明鹿茸蛋白提取物能提高环磷酰胺致免疫活性低下小鼠的单核巨噬细胞吞噬功能、血清溶菌酶浓度、血清及肝组织中酸性磷酸酶活力(P<0.05)。结论:本工艺条件可有效提高鹿茸蛋白提取率,且鹿茸提取物有良好的免疫活性。Objective: To optimize the best process of extracting soluble protein of Cervi Corun Pantotrichum by the aid of colloid mill and to verify the immune activity of the extract. Methods: Using Cervi Corun Pantotrichum protein extraction rate as an index, the effects of factors such as colloidal grinding teeth gap, colloid mill time, enzyme dosage,enzymolysis temperature, and enzymolysis time on protein extraction rate were studied by single factor experiments.Orthogonal experimental design was used to optimize the extraction process conditions and its immune activity was determined by immunological activity tests. Results: The optimum extraction process of colloid mill-assisted enzymeextracted soluble protein from Cervi Corun Pantotrichum was 50 μm for colloidal grinding teeth gap, 30 min for colloid mill time, 40 mg/g for enzyme dosage, 40 min for enzymolysis time, and 50 ℃ for enzymolysis temperature. The in vivo immune activity test showed that the Cervi Corun Pantotrichum protein extract could increase the phagocytic function of mononuclear macrophage, the concentration of serum lysozyme, and the activity of acid phosphatase in serum and liver tissue in mice with poor immune activity induced by cyclophosphamide(P<0.05). Conclusion: The technological conditions can effectively improve the extraction rate of Cervi Corun Pantotrichum protein, and the Cervi Corun Pantotrichum extract has good immune activity.
