摘 要:牛蒡子苷元(Arctigenin,ARG)是一种重要的广谱抗肿瘤药物。为利用微生物转化法制备牛蒡子苷元,以牛蒡子粉为底物以黑曲霉(Aspergillus niger)和里氏木霉(Trichoderma reesei)为菌种,混菌发酵转化合成牛蒡子苷元,利用高效液相色谱(High Performance Liqued Chromatography,HPLC)分析测定牛蒡子苷元。通过单因素试验对20L发酵罐进行最佳条件研究,结果表明:接种量为15%、发酵温度为29℃、搅拌转速为350r/min、通气量为1.0VVM。在单因素试验的基础上,采用Box-Behnken中心组合试验原理设计响应面方法优化工艺条件,得到最优发酵工艺条件为:接种量为14%、发酵温度为29℃、搅拌转速为335r/min、通气量为0.9VVM。在此条件下,牛蒡子苷元增加量为17.24mg/mL、牛蒡子苷转化率为98.7%。研究结果为微生物发酵转化合成牛蒡子苷元提供理论和实践基础,拓展了天然药用植物开发的路径。Arctigenin(ARG)is an important broad-spectrum anti-tumor drug.In order to prepare arctigenin by microbial transformation,arctigenin was synthesized by fermentation of mixture of Aspergillus Niger and Trichoderma Reesei with arctigenin powder as substrate,determination of arctigenin by High Performance liquid Chromatography(HPLC).The optimum conditions of 20L fermentor were studied by single factor experiment.The results showed that the inoculation amount was 15%,the fermentation temperature was 29℃,the agitation speed was 350r/min,and the aeration rate was 1.0VVM.On the basis of single factor test,the response surface method was used to optimize the process conditions by using the Box-Behnken central composite test principle,the optimal fermentation conditions were as follows:inoculation amount was 14%,fermentation temperature was 29℃,agitation speed was335r/min,aeration volume was 0.9VVM.Under these conditions,the content of Arctigenin increased to 17.24mg/mL,and the conversion rate of Arctigenin was 98.7%.The results provide theoretical and practical basis for the microbial fermentation of arctigenin,and expand the path of natural medicinal plants development.
