摘 要:目的:对外壳蛋白ε与非小细胞肺癌的关系、唾液酸化免疫球蛋白G的结合及其与肺癌的关系进行探索。方法:通过细胞照射、免疫沉淀联合质谱分析研究唾液酸化免疫球蛋白与外壳蛋白ε的结合;依托TCGA、GEO、GTEx、HPA、EGA及KM plotter公共数据库数据,使用R对外壳蛋白ε与非小细胞肺癌的关联及机制进行分析。结果:外壳蛋白ε与唾液酸化免疫球蛋白G结合且在射线照射后升高,在非小细胞肺癌中高表达并与分期和不良预后相关,其与多种促肿瘤通路正相关并与多种抗肿瘤通路负相关。结论:外壳蛋白ε与唾液酸化免疫球蛋白相互作用,并在非小细胞肺癌中发挥促肿瘤效应。Objective:No systematic reports were found onassociation between coatomer subunitεand nonsmall cell lung cancer.This study explored further on binding of sialylated-immunoglobulin-G to coatomer subunitεand the role of coatomer subunitεin lung cancer.Methods:Binding of sialylated-immunoglobulinG to coatomer subunitεwas validated by immunoprecipitation and liquid mass spectrometry after irradiation.R was applied for analysis of associations between coatomer subunitεand non-small cell lung cancer,as well as the possible mechanisms underlying,with database TCGA,GEO,GTEx,HPA,EGA and KM plotter.Results:Coatomer subunitε,binding to sialylated immunoglobulin G and increasing after irradiation,was highly expressed in non-small cell lung cancer and was associated with staging and poor prognosis.Multiple tumour promoting pathways were found positively correlated with coatomer subunitεwhile multiple tumour suppressing pathways negatively correlated.Conclusion:Coatomer subunitεinteracts with sialylated-immunoglobulin-G and acts as a tumour promoting factor in non-small cell lung cancer.
