摘 要:目的以秀丽隐杆线虫为模式生物,研究锁阳乙酸乙酯提取物(ECS)对衰老的影响和可能的作用机制。方法先设置空白组和ECS 0.1 mg/mL组、ECS 1 mg/mL组、ECS 5 mg/mL组、ECS 10 mg/mL组进行急性热应激实验初步筛选,然后在初筛的范围附近设置空白组和ECS 0.1 mg/mL组、ECS 0.4 mg/mL组、ECS 0.8 mg/mL组,分别进行急性热应激实验、寿命实验、生殖实验和运动能力实验,观察ECS对线虫的作用,结合转录组测序预测其可能的作用机制。结果急性热应激实验:ECS 1 mg/mL组、ECS 5 mg/mL组、ECS 10 mg/mL组线虫存活时间均明显短于空白组(P均<0.05),而ECS 0.1 mg/mL组、ECS 0.4 mg/mL组、ECS 0.8 mg/mL组线虫存活时间与空白组比较差异均无统计学意义(P均>0.05)。寿命实验:与空白组比较,ECS 0.1 mg/mL组线虫的存活时间无明显变化(P>0.05),ECS 0.4 mg/mL组线虫的存活时间明显延长(P<0.05),ECS 0.8 mg/mL组线虫的存活时间明显缩短(P<0.05)。生殖实验:各组产卵量比较差异均无统计学意义(P均>0.05)。运动能力实验:培养3 d和5 d后,ECS 0.4 mg/mL组线虫的头部摆动速率和身体正弦运动速率均明显高于空白组(P均<0.05)。KEGG分析富集排名前20的Pathway共富集到71个差异基因,包括超氧化物歧化酶(SOD)、硬脂酰辅酶A去饱和酶(SCD)、脂肪酶(LIPS)、热休克蛋白(HSP)等基因,涉及长寿调节通路、溶酶体途径、脂肪酸代谢通路、MAPK信号通路等多条代谢通路,参与调控ECS影响线虫寿命和运动能力的过程。结论ECS能够在一定程度上延长秀丽隐杆线虫的寿命,改善其运动能力,其是通过多途径多基因共同作用起效的。Objective Caenorhabditis elegans was used as a model organism to study the effect of ethyl acetate extract of Cynomorium songaricum(ECS)on aging and its possible mechanism.Methods The appropriate concentration range was preliminarily scre ened by acute heat stress test,which was divided into blank group,ECS 0.1 mg/mL group,ECS 1 mg/mL group,ECS 5 mg/mL group and ECS 10 mg/mL group,and then a new experimental group was set near the scope of preliminary screening,which was divided into blank group,ECS 0.1 mg/mL group,ECS 0.4 mg/mL group and ECS 0.8 mg/mL group for acute heat stress test,life span experiment and exercise ability experiment,to observe the effect of ECS on nematodes,and combined with transcriptome sequencing to predict its possible mechanism.Results Acute heat stress experiment:the survival time of nematodes of ECS 1 mg/mL group,ECS 5 mg/mL group and ECS 10 mg/mL group were significantly shorter than that of blank group(all P<0.05),there was no significant difference in the survival time among ECS 0.1 mg/mL group,ECS 0.4 mg/mL group and ECS 0.8 mg/mL group(all P>0.05).Life span experiment:compared with the blank group,the survival time of nematodes in ECS 0.1 mg/mL group was not significantly improved(P>0.05),the survival time of ECS 0.4 mg/mL group was significantly increased(P<0.05),and that of ECS 0.8 mg/mL group was significantly shortened(P<0.05).Reproductive experiment:there was no significant difference in oviposition among the groups(P>0.05).Exercise ability experiment:after 3 days and 5 days of ECS administration,the head swing rate and body sinusoidal motion rate of the ECS 0.4 mg/mL group were significantly higher than that of blank group(P<0.05).KEGG analysis enriched 71 differential genes in the top 20 pathways,including superoxide dismutase(SOD2),stearoyl COA desaturase(SCD),lipase(LIPS),heat shock protein(HSP)and other genes,involving multiple metabolic pathways such as longevity regulation pathway,lysosomal pathway,fatty acid metabolism pathway and MAPK signal pathway,which were involv
