摘 要:目的:探讨补肾活血方对尿酸性肾病大鼠肾功能及肾组织病理改变的影响,以及其对尿酸性肾病肾脏损伤的保护机制。方法:健康雄性SD大鼠予腺嘌呤和乙胺丁醇灌胃21 d,制备尿酸性肾病大鼠模型。将成模大鼠随机分为模型组(13只)、非布司他组(12只)、补肾活血方组(12只)、补肾活血方加非布司他组(12只)4组,另取12只空白鼠作为正常组。补肾活血方组每天予4 g/kg补肾活血方,非布司他组每天予非布司他水溶液3.6 mg/kg,补肾活血方加非布司他组予4 g/kg补肾活血方加3.6 mg/kg非布司他水溶液。于灌胃第2、4周各组处死6只大鼠,腹主动脉取血,测血尿酸、血肌酐、尿素氮。取肾脏组织,通过苏木素-伊红(Hematoxylin-eosin,HE)染色观察肾脏组织形态。结果:与正常组比较,模型组肾组织出现明显的病理改变;与正常组相比,模型组2周及4周血尿酸水平显著升高(P<0.01,P<0.05);2周时模型组尿素氮水平明显升高(P<0.05)。与模型组比较,非布司他组和补肾活血方组肾脏病理损伤明显减轻;与模型组相比,2周时非布司他组、补肾活血方加非布司他组血尿酸、尿素氮水平明显下降(P<0.05);4周时补肾活血方组、非布司他组、补肾活血方加非布司他组血尿酸水平明显下降(P<0.05)。结论:补肾活血方能够改善尿酸性肾病大鼠模型肾功能,减轻尿酸性肾病大鼠的肾脏损伤。Objective:To investigate the influences of the Bushen Huoxue prescription(补肾活血方)on renal function and pathological changes of renal tissue in rats with uric acid nephropathy,so as to explore its protective mechanism on renal injury in uric acid nephropathy.Methods:Healthy male SD rats were given adenine and ethambutol by gavage for 21 days to prepare rat models of uric acid nephropathy.The model rats were randomly divided into 4 groups,namely the model group(with 13 rats),the febuxostat group(with 12 rats),the Bushen Huoxue prescription group(with 12 rats),and the Bushen Huoxue prescription plus febuxostat group(with 12 rats),and another 12 blank rats were taken as the normal group.The model rats in the Bushen Huoxue prescription group were given The Bushen Huoxue prescription,4 g/kg per day,and the model rats in the febuxostat group were given febuxostat aqueous solution,3.6 mg/kg per day.The Bushen Huoxue presciption plus febuxostat group was given the Bushen Huoxue prescription plus febuxostat.At the 2nd and 4th week after intragastric administration,a total of 6 rats in each group were sacrificed.Blood was taken from the abdominal aorta.Blood uric acid,blood creatinine and urea nitrogen levels were measured,and kidney tissues were collected.The kidney tissue morphology was observed by HE staining.Results:Compared with the normal group,there were obvious pathological changes in the kidney tissue in the model group.Compared with the normal group,the blood uric acid level in the model group was significantly increased at the 2th week and 4th week(P<0.01,P<0.05),and the urea nitrogen level at the 2th week in the model group was significantly increased(P<0.05).Compared with the model group,the renal pathological damage in the febuxostat group and the Bushen Huoxue prescription group was significantly reduced.Compared with the model group,the levels of blood uric acid and urea nitrogen in the febuxostat group and the Bushen Huoxue prescription plus febuxostat group were significantly decreased at the 2th
