摘　　要：目的 探讨缺血性脑卒中(CIS)患者血清中Klotho蛋白的表达及其临床意义。方法 采用RT-qPCR,Western Blotting的方法检测2020年4月至2022年4月西安市西京医院收治的52例CIS患者及52例正常对照组的血清中Klotho的表达,并分析其与患者疾病活动度的相关性。结果 Klotho的mRNA在CIS患者血清标本中的相对表达量低于正常对照组,差异有统计学意义(t=17.986,P<0.05);Western Blotting检测显示,Klotho蛋白在CIS血清标本中相对表达量低于正常对照组,差异有统计学意义(t=15.554,P<0.05);Klotho蛋白表达水平与CIS患者疾病活动度NIHSS评分呈负相关关系(r=-0.5974,P<0.05),与BI评分呈正相关关系(r=0.6069,P<0.05)。结论 Klotho可能参与缺血性脑卒中的发生发展。Objective To investigate the expression and clinical significance of Klotho protein in serum of patients with CIS. Methods RT-qPCR and Western blotting were used to detect the expression of Klotho in serum of 52 patients with CIS and 52 normal control groups in Xi’an Xijing Hospital from April 2020to April 2022. The correlation between the expression of Klotho and disease activity of the patients was analyzed.Results The relative expression of Klotho m RNA in serum samples of CIS patients was lower than that of normal control groups,and the difference was statistically significant(t=17.986,P<0.05). The Western blotting results showed that the protein expression of Klotho in serum samples of patients with CIS were lower than that of the normal control group,with statistically(t=15.554,P<0.05). The protein expression of Klotho was negatively correlated with the NIHSS score of disease activity in patients with CIS(r=-0.5974,P<0.05)and positively correlated with the BI score of disease activity in patients with CIS(r=0.6069, P<0.05).Conclusion Klotho may be involved in the occurrence and development of ischemic stroke.
