摘  要:研究了抑制性受体FcγRIIB在肝细胞癌(HCC)中的表达,为探讨肝脏炎癌转化的调控机制提供理论基础.利用肿瘤基因组图谱(TCGA)、基因表达综合数据库(GEO)和肿瘤数据库(Oncomine)分析肝癌中FcγRIIBmRNA的表达,采用实时定量PCR(qRT-PCR)验证FcγRIIB在临床HCC患者肿瘤样本中的表达,通过单次注射N-亚硝基二乙胺(DEN)诱导小鼠HCC并检测FcγRIIB的表达.生物信息学分析显示FcγRIIB在HCC中低表达;qRT-PCR结果显示,临床HCC患者肿瘤样本和DEN诱导小鼠肝癌模型中FcγRIIB的表达均降低.生物信息学、临床样本和小鼠模型三方面得出一致结论,即FcγRIIB在HCC中低表达,提示FcγRIIB可能成为肝癌诊断或预后新的生物标志物,并在肝脏炎癌转化中发挥重要作用.This study is performed to assess the expression of inhibitory receptor FcγRIIB in HCC, which provides the theoretical basis for exploring the regulatory mechanism of hepatic inflammation-induced carcinogenesis. The expression of FcγRIIB mRNA in HCC was analyzed by TCGA, GEO and Oncomine databases and verified in tumor samples of clinical HCC patients by qRT-PCR. HCC in mouse was induced by a single injection of DEN, and the expression of FcγRIIB was detected. Bioinformatics analysis showed that the expression of FcγRIIB was low in HCC. qRT-PCR results showed that the expression of FcγRIIB decreased in tumor samples of clinical HCC patients and DEN-induced mouse HCC model. The consistent conclusion from bioinformatics, clinical samples and mouse model was that the expression of FcγRIIB was low in HCC. These data suggest that FcγRIIB may become a new biomarker for the diagnosis or prognosis of liver cancer and play an important role in the hepatic inflammation-induced carcinogenesis.
