摘  要:目的:研究制备青花椒精油纳米乳的最优配方,并对其理化性质和体外抑菌活性进行考察。方法:采用相转变法制备青花椒精油纳米乳并绘制伪三相图筛选其配方工艺,然后使用粒径分析仪、透射电子显微镜等对其表征形态进行检测,最后考察青花椒精油纳米乳在体外对大肠杆菌和金黄色葡萄球菌的抑制活性。结果:青花椒精油纳米乳最优配方为:12.63%蓖麻油聚氧乙烯醚(EL-40)、4.21%1,2-丙二醇、7.33%青花椒精油、75.82%蒸馏水。以此配方为基础制备的青花椒精油纳米乳整体澄清透明、流动性和稳定性均良好。试验制备的青花椒精油纳米乳平均粒径为126.1±0.78 nm,分散指数为0.591±0.21,呈正态分布;透射电镜下其形态为球形,分布均匀、分散性良好,载药量为73.29±1.56 mg/mL。体外抑菌实验表明,青花椒精油纳米乳对金黄色葡萄球菌和大肠杆菌的MIC值均为2.29 mg/ml,且青花椒精油纳米乳抑菌效果优于青花椒精油。结论:制备得到的青花椒精油纳米乳工艺简单,显著提升了青花椒精油的抑菌活性。Objective:The optimum formulation of Zanthoxylum essential oil nanoemulsion was studied,and its physical and chemical properties and antibacterial activity in vitro were investigated.Method:Zanthoxylum essential oil nanoemulsion was prepared by phase transformation method and its formulation process was screened by pseudo-three-phase diagram.Then,the characterization morphology of Zanthoxylum essential oil nanoemulsion was detected by particle size analyzer and transmission electron microscope.Finally,the inhibitory activity of Zanthoxylum essential oil nanoemulsion against Escherichia coli and Staphylococcus aureus in vitro was investigated.Results:The optimal formula of Zanthoxylum essential oil nanoemulsion was 12.63%castor oil polyoxyethylene ether(EL-40),4.21%1,2-propylene glycol,7.33%Zanthoxylum essential oil and 75.82%distilled water.The Zanthoxylum essential oil nanoemulsion prepared based on this formula had good clarity,transparency,fluidity and stability.In this experiment,the average particle size of prickly ash essential oil nanoemulsion prepared was 126.1±0.78 nm,and the dispersion index was 0.591±0.21,showing normal distribution.Under transmission electron microscope,it was spherical in shape,evenly distributed and well dispersed,and the drug loading was 73.29±1.56 mg/mL.In vitro antibacterial experiments showed that the MIC values of Zanthoxylum essential oil nanoemulsion against Staphylococcus aureus and Escherichia coli were 2.29mg/mL,and the antibacterial effect of Zanthoxylum essential oil nanoemulsion was better than that of Zanthoxylum essential oil.Conclusion:The Zanthoxylum essential oil nanoemulsion prepared in this study had a simple process,which significantly improved the antibacterial activity of Zanthoxylum essential oil.
