摘 要:为分析折耳根(蕺菜)提取物对番茄灰霉病菌的抑制作用,采用番茄活体接种法(以4叶1心的番茄幼苗为试验材料)和培养基接种法(体外抑菌试验),分析比较折耳根提取液对灰霉病菌的抑菌作用。番茄活体接种试验结果表明:折耳根提取液预处理的叶片发病程度比蒸馏水处理(对照)的轻,在接种病害3 d后,发病率和病情指数分别下调至81.5%和3.0左右,灰霉Actin基因相对表达量显著低于对照处理,叶片叶绿素a、b含量分别为4.96(对照的1.25倍)、2.23 mg/L(对照的1.30倍),均显著高于对照;折耳根提取液预处理和接种灰霉病菌均导致番茄叶片脂氧合酶(LOX)活性上升,接种灰霉菌后,折耳根提取液处理的叶片LOX活性为13.52 OD/g(FW),显著高于蒸馏水处理(对照)的10.92 OD/g(FW),且两者均显著高于未接种灰霉病菌的预处理。体外抑菌试验表明:折耳根提取液处理后灰霉菌的长势及扩散程度小于蒸馏水处理的对照,表明其对番茄体外培养的灰霉病菌生长也有抑制作用。In order to analyze whether the extract of Euphorbia pulcherrima had inhibitory effect on Botrytis cinerea,in vivo inoculating method(tomato seedlings with four leaves and one heart as material)and culture medium inoculating method(in vivo bacteriostasis test)of tomato were used to analyze and compare the effect of Houttuynia cordata extract on inhibition of Botrytis cinerea of tomato.The results of in vivo inoculation test showed that the leaf pathogenicity of the pretreated extract was lighter than that of the water treatment control.After 3 d inoculation,the incidence and disease index of the group were reduced to 81.5%and 3.0,respectively,the relative expression of grey mould Actin in tomato leaves pretreated with follicle extract was significantly lower than that of the control,and the contents of chlorophyll a and chlorophyll b in leaves were 4.96(1.24 times of the control)and 2.23 mg/L(1.3 times of the control),respectively,which were significantly higher than that of the control.Lipoxygenase(LOX)activity of tomato leaves increased by the treatment of the extract of Hotrytis cinerea and inoculation of Botrytis cinerea.After inoculation of Botrytis cinerea,LOX activity of the leaves treated with the extract of Hotrytis cinerea was 13.52 OD/g(FW),significantly higher than that of the control treatment of 10.92 OD/g(FW),and both were significantly higher than that of the pretreatment without Botrytis cinerea.The in vitro bacteriostasis test showed that the growth and diffusion of Botrytis cinerea after the treatment of the extract of Hotrytis cinerea were less than those of the control treated with distilled water,indicating that it also had an inhibitory effect on the growth of Botrytis cinerea in vitro.
