circ_0046263靶向miR-1184调控肝癌HCCLM3细胞增殖、迁移和侵袭的机制研究

摘　　要：目的:探讨circ_0046263对肝癌HCCLM3细胞增殖、迁移和侵袭的影响及分子机制。方法:选取惠州市第三人民医院2017年1月至2020年12月收治的47例肝癌患者肿瘤组织及癌旁组织,RT-qPCR检测circ_0046263和miR-1184表达;将肝癌细胞株HCCLM3随机分为si-NC组、si-circ_0046863组、miR-NC组、miR-1184组、si-circ_0046263+anti-miR-NC组、si-circ_0046263+anti-miR-1184组,CCK-8检测细胞活性;克隆形成实验检测细胞克隆形成数;划痕实验检测划痕愈合率;Transwell检测侵袭细胞数;Western blot检测蛋白表达;双荧光素酶报告实验检测circ_0046263和miR-1184的靶向关系。结果:与癌旁组织相比,肝癌组织circ_0046263表达升高,miR-1184表达降低(P<0.05)。抑制circ_0046263表达或过表达miR-1184后,肝癌细胞HCCLM3活性降低,细胞克隆形成数减少,细胞划痕愈合率降低,侵袭细胞数减少,E-cadherin表达升高,N-cadherin表达降低(P<0.05)。circ_0046263靶向调控miR-1184表达。下调miR-1184表达逆转了抑制circ_0046263表达对肝癌HCCLM3细胞增殖、迁移和侵袭的作用。结论:抑制circ_0046263表达通过上调miR-1184表达抑制肝癌HCCLM3细胞增殖、迁移和侵袭。Objective:To explore effect of circ_0046263 on proliferation,migration and invasion of liver cancer HCCLM3cells and its molecular mechanism.Methods:A total of 47 cases of liver cancer tissues and adjacent tissues of liver cancer patients in Huizhou Third People′s Hospital from January 2017 to December 2020 were selected;RT-qPCR was used to detect expressions of circ_0046263 and miR-1184;liver cancer cell line HCCLM3 was randomized divided into si-NC group,si-circ_0046863 group,miR-NC group,miR-1184 group,si-circ_0046263+anti-miR-NC group,si-circ_0046263+anti-miR-1184 group;CCK-8 was used to detect cell viability;clone formation test was used to detect cell clone formation number;scratch test was used to detect scratch healing rate;Transwell was used to detect number of invaded cells;Western blot was used to detect protein expressions;dual luciferase report experiment was used to detect targeting relationship between circ_0046263 and miR-1184.Results:Compared with adjacent tissues,expression of circ_0046263 in liver cancer tissues was increased,miR-1184 expression was decreased(P<0.05). After inhibiting expression of circ_0046263 or overexpressing miR-1184,activity of hepatocellular carcinoma HCCLM3 cells was decreased,number of cell clone formation was decreased,healing rate of cell scratches was decreased,number of invasive cells was decreased,E-cadherin expression was increased,N-cadherin expression was decreased(P<0.05). circ_0046263 targeting regulated miR-1184 expression;down-regulation of miR-1184 expression reversed effect of inhibiting circ_0046263 expression on proliferation,migration and invasion of HCCLM3 cells.Conclusion:Inhibition of expression of circ_0046263 can inhibit proliferation,migration and invasion of HCCLM3 cells by up-regulating miR-1184 expression.
