摘　　要：目的:探讨lncRNA DICER1-AS1对肺癌细胞增殖、迁移的影响及分子机制。方法:收集2018年1月至2020年1月杭州市萧山区中医院23例肺癌和癌旁组织标本,采用RT-qPCR检测lncRNA DICER1-AS1和miR-206的表达水平;将A549细胞随机分为si-NC组、si-lncRNA DICER1-AS1组、si-lncRNA DICER1-AS1+anti-miR-NC组、si-lncRNA DICER1-AS1+anti-miR-206组;RT-qPCR检测各组细胞中lncRNA DICER1-AS1和miR-206的表达水平;MTT法检测细胞存活率;克隆形成实验检测细胞克隆形成数;Transwell检测细胞迁移;Western blot法检测蛋白表达;荧光素酶报告实验检测lncRNA DICER1-AS1和miR-206的靶向关系。结果:肺癌组织中lncRNA DICER1-AS1呈高表达,miR-206呈低表达(P<0.05)。荧光素酶报告实验显示lncRNA DICER1-AS1直接靶向调控miR-206。干扰A549细胞中lncRNA DICER1-AS1表达,miR-206表达水平升高,细胞存活率降低,克隆形成数减少,迁移细胞数减少,Ki67、N-cadherin表达水平降低,E-cadherin表达水平升高(P<0.05)。抑制miR-206表达可逆转干扰lncRNA DICER1-AS1对肺癌细胞增殖、迁移的抑制作用。结论:干扰lncRNA DICER1-AS1可能通过上调miR-206表达抑制肺癌细胞增殖、迁移。Objective:To investigate the effect and molecular mechanism of lncRNA DICER1-AS1 on lung cancer cell proliferation and migration.Methods:Collected 23 cases lung cancer tissue and paracancerous tissue specimens from January 2018 to January 2020 in Xiaoshan Hospital of Traditional Chinese Medicine,RT-qPCR was used to detect the expression levels of lncRNA DICER1-AS1 and miR-206;A549 cells were randomly divided into si-NC group,si-lncRNA DICER1-AS1 group,si-lncRNA DICER1-AS1+anti-miR-NC group,si-lncRNA DICER1-AS1+anti-miR-206 group;RT-qPCR was used to detect the expression levels of lncRNA DICER1-AS1 and miR-206 in each group of cells;MTT was used to detect cell survival rate;clone formation was used to detect the number of cell clones formed;Transwell was used to detect cell migration;Western blot was used to detect protein expression;luciferase report was used to detect the targeting relationship between lncRNA DICER1-AS1 and miR-206.Results:lncRNA DICER1-AS1 was over-expressed in lung cancer tissues,and miR-206 was down-expressed(P<0.05). Interference with lncRNA DICER1-AS1 expression,miR-206 expression were increased,cell survival rate was decreased,A549 cell clone formation number was decreased,A549 migration cell number was decreased,Ki67 and N-cadherin expressions in A549 cells were decreased,and E-cadherin expression in A549 cells was increased(P<0.05). lncRNA DICER1-AS1 targets and regulates miR-206,inhibiting miR-206 expression can reverse the inhibitory effect of interfering with lncRNA DICER1-AS1 on the proliferation and migration of lung cancer cells.Conclusion:Interfering with lncRNA DICER1-AS1 may inhibit the proliferation and migration of lung cancer cells by up-regulating the expression of miR-206.
