摘 要:目的探讨水泡性口炎病毒(VSV)对结肠癌SW480细胞增殖、迁移和侵袭的影响及其分子机制。方法用Vero细胞对VSV进行扩增、浓缩和滴度测定。通过CCK-8法检测不同浓度的VSV对SW480细胞增殖活性的抑制作用;采用划痕实验、Transwell实验检测SW480细胞的迁移和侵袭能力;实时荧光定量聚合酶反应(qPCR)及蛋白质印迹法(Western blotting)检测SW480细胞的MMP-2、MMP-9 mRNA及蛋白表达。结果VSV处理组增殖率小于对照组,差异有统计学意义(P<0.05)。VSV处理组细胞的迁移能力低于对照组,差异有统计学意义(P<0.05)。VSV处理组侵袭细胞数明显低于对照组,差异有统计学意义(P<0.05)。VSV处理组MMP-2表达量高于对照组,MMP-9表达量低于对照组,差异有统计学意义(P<0.05)。结论VSV通过靶向调控基质金属蛋白的表达能够抑制结肠癌SW480细胞的侵袭、迁移。Objective To investigate the impacts of vesicular stomatitis virus(VSV)on the proliferation,migration,and invasion of colon cancer SW480 cells and its molecular mechanism.Methods Vero cells were used to amplify,concentrate,and determine the titer of VSV.CCK-8 method was used to detect the inhibitory effect of different concentrations of VSV on the proliferation activity of SW480 cells.Scratch test and Transwell test were used to detect the migration and invasion ability of SW480 cells.The expressions of MMP-2,MMP-9 mRNA and protein in SW480 cells were detected by real-time quantitative polymerase reaction(qPCR)and Western blotting.Results The proliferation rate of the VSV treatment group was lower than that of the control group,with statistically significant difference(P<0.05).The migration ability of cells in the VSV treatment group was lower than that in the control group,with statistically significant difference(P<0.05).The number of invasive cells in the VSV treatment group was significantly lower than that in the control group,with statistically significant difference(P<0.05).The expression of MMP-2 in the VSV treatment group was higher than that in the control group,and the expression of MMP-9 was lower than that in the control group,with statistically significant differences(P<0.05).Conclusion VSV inhibits the invasion and migration of colon cancer SW480 cells by targeting and regulating the expression of matrix meta lloproteins.
