摘  要:目的观察雪莲果浸膏对高脂模型大鼠的降脂作用及机制。方法利用网络药理学方法对雪莲果干预高脂血症的可能分子机制进行预测,确定脂质代谢研究对象;80只SD大鼠随机选取20只作为空白组(普通饲料饲养8周),60只作为高脂模型组(用高脂高糖饲料饲养8周建模);建模成功后高脂模型组大鼠再分为阳性对照组、模型组及雪莲果浸膏(高、中、低)剂量组,雪莲果浸膏3个剂量组给予相应浓度雪莲果浸膏灌胃、阳性对照组给予奥利司他灌胃、其余各组给予生理盐水灌胃,连续8周,观察各组大鼠的体质量变化;采用酶联免疫吸附法检测各组大鼠血清总甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)含量,实时荧光定量PCR及免疫印迹法分别检测各组大鼠胰腺组织中胰脂肪酶、辅脂酶的m RNA及蛋白水平。结果通过网络药理学对雪莲果干预高脂血症的可能分子机制进行预测分析,确定胰脂肪酶和辅脂酶为脂质代谢研究的目标;动物实验结果显示,与模型组比较,雪莲果浸膏可降低高脂大鼠胰腺组织中胰脂肪酶、辅脂酶m RNA及蛋白表达水平(P<0.05),能改善高脂模型大鼠的血脂情况。结论雪莲果浸膏具有降脂作用,其机制为抑制胰脂肪酶、辅脂酶表达,抑制胰腺中胰脂肪酶、辅脂酶的合成,降低大鼠肠道对脂质的吸收。Objective To observe the lipid-lowering effect of yacon extract on the pancreatic lipase and colipase in rats(rattus norvegicus)with high-fat model and explore its effect on lipid absorption in rats.Methods The network pharmacology method was used to predict the possible molecular mechanism of hyperlipidemia in Saussurea involucrata;lipase and lipid metabolism were selected as the research objects.A total of 80 SD rats were randomly selected as the blank group(fed with ordinary diet for 8 weeks)and the high-fat model group(fed with high-fat and high-sugar diet for 8 weeks).After the success of the modeling,the high-fat model group rats were divided into positive control group,model group and tuber concrete of high,medium,and low dose group.The three dose groups of tuber extract were given the corresponding concentration tuber extract lavage,and the positive control group was given orlistat lavage as well as the groups were given normal saline lavage,the weight of rats in each group was observed for 8 weeks.serum samples were collected to detect the total triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)by enzyme-linked immunosorbent assay(ELISA).The pancreatic lipase,colipase mRNA levels and protein content were detected by real-time quantitative PCR(qPCR)and Western blot,respectively.Results The possible molecular mechanism of Saussurea involucrata in the intervention of hyperlipidemia was predicted and analyzed by network pharmacology,and pancrelipase and colipase were identified as lipid metabolism research objects.The results of animal experiments showed that Saussurea extract could reduce the mRNA and protein expression levels of pancreatic lipase and colipase in the pancreatic tissue of high-fat rats compared with the model group(P<0.05),and could improve the lipid profile of high-fat rats.Conclusion The yacon extract has a lipid-lowering effect,which can inhibit the synthesis of pancreatic lipase and colipase in the pancreas of
