摘  要:目的 观察异氰酸烯丙酯对大鼠垂体腺瘤细胞(GH3细胞)生物活性的影响,并探讨其作用机制。方法 常规培养大鼠GH3细胞,将细胞随机分为0.1、1、10μmol/L实验组及对照组,分别给予终浓度0.1、1、10μmol/L异硫氰酸烯丙酯及等体积DMSO继续培养。用细胞增殖试验检测细胞活力,流式细胞术检测细胞周期,ELISA法检测细胞培养上清液中生长激素(GH)、胰岛素样生长因子-1(IGF-1),Western blotting法检测细胞内细胞周期相关蛋白[周期蛋白激酶-4(CDK-4)、p21、p27蛋白]表达。结果 培养24、48、72 h,对照组及0.1、1、10μmol/L实验组增殖能力(OD值)逐渐降低(P均<0.05),G1期细胞比例逐渐升高、S期细胞比例逐渐降低(P均<0.05),细胞培养上清液中GH、IGF-1水平逐渐降低(P均<0.05),CDK4蛋白相对表达量逐渐降低、p21和p27蛋白相对表达量逐渐升高(P均<0.05)。结论 异硫氰酸烯丙酯素可抑制GH3细胞生物活性,其机制可能与调控细胞周期相关蛋白(CDK4、p21、p27蛋白)表达有关。Objective To study the effect of allyl isocyanate(Ally) on the biological activity of pituitary adenoma GH3 cells and to investigate its mechanism.Methods Rat GH3 cells were routinely cultured,and were randomly divided into the control group and experimental group.GH3 cells in the experimental groups were treated with Ally(0.1μmol/L,1 μmol/Land 10 μmol/L),and GH3 cells in the control group were treated with equal volume of dimethlsulfoxide.Cell proliferation was measured by MTS experiment;flow cytometry was used to measure the cell cycle;ELISA was applied to measure the level of growth hormone(GH) and insulin growth factor-1(IGF-1) in cell culture supernatant;and Western blotting was used to detect the level of cell cycle-related protein [cyclin kinase 4(CDK4),p21 and p27 protein).Results After 24,48 and 72 h of culture,the cell proliferation ability(OD value) decreased gradually in the 0.1,1 and10 μmol/L experimental groups and control group(all P<0.05),the percentage of cells in the G_(1) phase increased gradually,and percentage of cells in the S phase decreased gradually(all P<0.05);the levels of GH and IGF-1 decreased gradually(all P<0.05);the relative expression levels of CDK protein gradually decreased;and the p21 and p27 increased gradually(all P<0.05).Conclusion Ally inhibits the biological activity of GH3 cells probably by regulating the expression levels of CDK4,p21 and p27 proteins.
